影響繍球孢子屬叢枝內生菌根菌孢子萌芽之因素

Factors on the Spores Germination of Arbuscular Mycorrhizal Fungi, Glomus Spp

摘要


本試驗將表面消毒之Glomus spp.孢子,置於1%水洋菜培養基上培養,探討溫度、pH值、培養基內添加物以及接種源(培養土)之貯存溫度與時間對孢子萌芽之影響。結果顯示G. mosseae(Gm)、G. clarum(Gc)及G. etunicatum(Ge)之孢子於28℃,G. fasciculatum (Gf)於20-24℃萌芽率最高。供試四菌種對pH值適應範圍廣,孢子在pH4-10均可萌發,但以pH7-8萌芽率較高。Gf孢子在水洋菜培養基萌芽率最高(42%),水洋菜培養基上添加根分泌物,可使Gm孢子萌芽率由24%提升至37%。收穫之Gm與Gf菌根菌乾燥培養土於室溫下貯存2個月,水洋菜培養基上孢子萌芽率分別為45%及69%;將之再分別貯存於室溫、4℃及-20℃,結果顯示收獲後培養土經室溫貯存8個月,Gm孢子萌芽率提升至68%,Gf提升至83%;但至第14個月時二者之孢子萌芽率低於4℃貯存者。於4℃貯存之培養土於第14個月,Gm及Gf孢子萌芽率亦較貯存於-20℃者高,是故接種源培養土短期(8個月內)貯存於室溫即可,長期貯存則以4℃為佳。

Spores of Glomus spp. were isolated and surface-sterilized before incubating on a 1% water agar to study the effects of temperature, pH, amended media, as well as the preservation temperature and duration of the inoculum (culture soil) on their germination. The results showed that the maximum spore germination occurred at 28℃ for G. mosseae (Gm), G. clarum (Gc), and G. etunicatum (Ge), 20-24℃ for G. fasciculatum (Gf). The pH experiment showed that all the 4 species of Glomus spores were able to germinate between pH4-10 and the optimal was 7-8. In the media experiment, water agar supported excellent spore germination of Gf (42%). Amending root exudate to water agar promoted the germination rate of Gm from 24% to 37%. Should the dry culture soils have been stored at room temperature for 2 months, the spore germination rate of the readily harvested Gm and Gf on water agar were raised to 45% and 69% respectively. If culture soils were stored at 4t, room temperature and -20℃ respectively, the spore germination rate of Gm would be promoted from 45% to 68%, Gf from 69% to 83% with room temperature preservation for 8 months. Both the spore germination rate of Gm and Gf declined after 14 months, and were lower than those having been stored at 4℃. Those stored at -20℃ also showed inferior to those at 4℃. Hence, room temperature was the optimal temperature for inoculum preservation within 8 months, while, 4℃ was better than room temperature and -20℃ for a longer preservation period.
