不同蕪菁嵌紋病毒系統之鞘蛋白質基因序列之比較

摘要


蕪菁嵌紋病毒(turnip mosaic virus;簡稱TuMV)屬於Potyvirus病毒屬,可以感染許多重要的經濟作物,因此被認為是世界上感染十字花科最重要的病毒之一。將六個病毒系統TuMV- C1、C2、C3、C4、C5和TW之芥菜病葉之萃取液,經PEG沈降及硫酸銫密度離心後可得到純化之病毒顆粒,病毒RNA萃取後經電泳顯示其長度均約10 kb。進一步以反轉錄聚合酵素連鎖反應選殖六個病毒系統之鞘蛋白質基因,譯讀出其核啟酸序列,並比較六者之鞘蛋白質基因核啟酸序列、胺基酸序列與3'端非轉譯區核啟酸序列之相同度。結果顯示六者之間鞘蛋白質核啟酸相同度高達92.7∼99.3%,胺基酸相同度高達93.8∼99.0%,3'端非轉譯區核啟酸相同度則高達98.9∼100%且含有約62%的AU。比較這六個TuMV病毒系統與國外報告之病毒系統CPP(中國大陸),CAPP(加拿大),CQS、NIAP(韓國),CP、CPRN、MUT(英國)和JAP(日本)等,在鞘蛋白質基因之核啟酸序列、胺基酸序列與3'端非轉譯區核啟酸序列之相同度,結果顯示,TuMV- C5、TW與韓國NIAP系統較為接近。六個系統中僅TuMV- C1、C5和TW和大部分國外不同地區系統之鞘蛋白胺基酸位於第6∼8胺基酸位置具有與蚜蟲傳播有關DAG結構。由於此六個TuMV系統在血清學上不易區分,為了發展有效對抗TuMV的防治策略和分子診斷技術,分析此一病毒的遺傳差異性是有其必要性。

TuMV is a member of the genus potyvirus. It infects many economically important crops, and is one of the most important viruses in the world affecting Brassica species. In this study virons of six TuMV strains, C1、C2、C3、C4、C5 and TW, were purified from infected leaves of mustard by polyethylene glycol precipitation followed by a Cs_2SO_4 isopycnic centrifugation. Electrophoresis revealed presence of a single RNA band about 10 kb for each strain of TuMV. The cDNAs of the coat protein (CP) genes and 3'nontranslated regions of the six TuMV strains were synthesized by reverse transcription polymerase chain reaction (RT-PCR). Analysis of nucleotide sequences and amino acid sequences of the CP genes and the 3' non-translated regions indicated that the CP genes of TuMV- C1、C2、C3 and C4, and those of TuMV- C 5 and TW share close relationships. The CP genes and 3' non-coding regions of the six TuMV strains were also compared with other reported TuMV strains of CPP (from China), CAPP (from Canada), CQS, NIAP (from Korea), CP, CPRN, MUT (from UK) and JAP (from Japan). Results showed that the CP genes of Tu M V-C5 and TW have closer relationships with the NIAP strain from Korea.
