台灣紅豆杉扦插苗接種農桿根群菌對苗木生長與紫杉烷類含量之影響

Growth Performance and Taxane Content of Taxus mairei Cuttings with Roots Induced by Agrobacterium rhizogenes

摘要


採自台灣紅豆杉成熟母樹之插穗,經由對照組、3000 ppm IBA與3種農桿根群菌(R1600,R1601,R15834)等三種誘導發根處理後,將此5種處理苗木栽植於裝填有蛭石、泥炭土與肥料的混合介質生長9.5月後,調查幼苗的生長表現與其紫杉烷類含量。農桿根群菌處理苗在初植時的根數是對照組的2~3倍,IBA處理苗的1.1~1.5倍。經生長9.5月後,R1600處理苗的乾物重最大,而對照組苗最小。相關分析顯示苗木初植的根數與植物生長呈現極顯著正相關,而與苗木初植的重量無關。5種處理苗木間的紫杉醇濃度差異不顯著,但10-deacetylbaccatin Ⅲ (10-DAB)則顯示有顯著差異。生物量最大的R1600處理苗與最小的對照組苗濃度最低,不過由整株4種處理苗萃取10-DAB的收益間差異不顯著,但處理苗與對照組苗間呈顯著差異。

Root formation of cuttings collected from a Taxus mairei adult tree was induced by treatment with 0 (control) and 3000 ppm IBA, and 3 strains of Agrobacterium rhizogenes (R1600, R1601, and R15834). Root numbers of plants treated with Agrobacterium were 2~3 times greater than those of control plants and 1.1~1.5 times greater than those of IBA-treated plants. These plants were planted into pots containing a mixture of vermiculite, peat, and fertilizer for 9.5 mo to determine the growth performance and taxane contents. R1600-treated plants had the greatest dry weight, while the control plants had the smallest. Correlation analysis indicated that root numbers of transplanted plants were positively related to plant biomass, but not to initial plant weight. Taxol concentrations among the 5 treatments did not significantly differ, while 10-deacetylbaccatin (10-DAB) contents did. The treated plants (R1600 and control) with the greatest and least mass respectively had lower concentrations. However, no significant differences in yields of 10-DAB extracted from whole plants between IBA-and Agrobacterium-treated plants were found, but there was a significant difference between treated and control plants.
