市售常見中草藥植物抗氧化活性之評估

Evalution of antioxidant activity on commercial common herbal medicine plants

摘要


本研究之目的乃使用市售常見的9種中草藥植物,以水或95%乙醇萃取,進行其萃取物抗氧化能力及活性之評估。分析項目包括:總酚化合物含量、超氧歧化酶(superoxide dismutase, SOD)活性、清除DPPH(1, l-diphenyl-2-picryl hydrazyl)自由基能力及總抗氧化能力。結果顯示,9種中草藥植物樣品萃取物之總酚化合物含量,其值介於24.17-713.04 mg GAE/g之間,含量前3高者,依序為虎杖乾燥品(乙醇萃)713.04 mg GAE/g、大葉骨碎補乾燥品(乙醇萃)643.56 mg GAE/g及大葉骨碎補鮮品凍乾品(乙醇萃)573.40 mg GAE/g。SOD活性方面,其活性值範園為0-936.76 U/mg 間, 在全部樣品中SOD活性前3高者,依序為普洱茶(大陸雲南)(乙醇萃)936.76 U/mg、大葉骨碎補乾燥品(乙醇萃) 355.96 U/mg及大葉骨碎補鮮品凍乾品(乙醇萃)259.11 U/mg。清除DPPH自由基能力方面,雖然每種樣品萃取物清除DPPH自由基能力高低不一,但每種樣品皆呈劑量效應,隨著濃度的提高而增加,清除DPPH自由基能力最佳之前3者,當濃度為50 μg/mL時,依序為大葉骨碎補鮮品凍乾品(乙醇萃)61.54%、虎杖乾燥品(乙醇萃)51.35%及普洱茶(大陸雲南)(水萃)為47.63%。總抗氧化能力方面,每種樣品皆呈劑量效應,也隨濃度提高而增加,於低濃度下即顯示有良好清除ABTS^(.+)自由基之能力,當濃度為5 0 μg/mL時,清除ABTS^(.+)自由基能力前3高者,依序為普洱茶(大陸雲南)(水萃)54.49%、虎杖乾燥品(乙醇萃)43.71%及大葉骨碎補鮮品凍乾品(乙醇萃)為40.25%。由以上結果顯示市售常見9種中草藥植物雖抗氧化成分及清除自由基的能力結果不一,各項中草藥萃取物樣品於低濃度下,即可發現有較高的抗氧化效應,因此推測中草藥類植物仍具有較佳的抗氧化能力。

There were 9 kinds of commercial common herbal plants used in this study. The ways of extraction were used water or 95% ethanol, and evaluated the antioxidant capacity and activity of various samples. Analyzing items included total phenolic contents, superoxide dismutase activity (SOD), DPPH radical scavenging activity and total antioxidant capacity. The results indicated that the content of phenolic compounds in various sample extracts were between 24.17-713.04 mg GAE/g. The top three of contents of total phenolic compounds were 713.04 mg GAE/g of dried Polygonum cuspidatum Sieb. et Zucc (ethanolic extracts), 643.56 mg GAE/g of dired Davallia mariesii Moore (ethanolic extracts) , and 573.40 mg GAE/g of fresh and freezed Davallia mariesii Moore (ethanolic extracts), respectively. The SOD activity of herbal plants were 0- 936.76 U/mg. The top three of SOD activity in all samples were 936.76 U/mg of Pu-Erh tea (China Yunnan) (ethanolic extracts), 355.96 U/mg of dired Davallia mariesii Moore (ethanolic extracts), and 259.11 U/mg of fresh and freezed Davallia mariesii Moore (ethanolic extracts), respectively. In DPPH radical scavenging activity, despite the fact that the scavenging activity in each sample extract was different, it still found that all the samples had the dose response and enhanced with increase of concentration. The top three of commercial common herbal plants, when the concentration was at 50 μg/mL, the DPPH radical scavenging activity of fresh and freezed Davallia mariesii Moore (ethanolic extracts) was 61.54%, dried Polygonum cuspidatum Sieb. et Zucc (ethanolic extracts) was 51.35%, and Pu-Erh tea (China Yunnan) (water extracts) was 47.63%. In total antioxidant activity, all the samples showed the dose response and enhanced with increase of concentration. The top three of total antioxidant activity, which had good ABTS^(.+) free radical scavenging activity with a low concentration. When the concentration was at 50 μg/ml., the ABTS^(.+) free radical scavenging activity of Pu-Erh tea (China Yunnan) (water extracts) was 54.49%, dried Polygonum cuspidatum Sieb. et Zucc (ethanolic extracts) was 43.71 %, and fresh and freezed Davallia mariesii Moore ethanolic extracts) was 40.25%, respectively. From the above results show that the 9 kinds of common herbal plants which their antioxidant composition and the ability of scavenge free radicals were different , and that could be found a better antioxidant response with a low concentration. Thus, the study surmised that herba l plants had the better antioxidant activity.
