高分子量幾丁聚醣對餵食高脂飼料大白鼠體重、營養素消化率及血脂質之影響

Effect of High-molecular-weight Chitosan on Body Weight, Nutrient Digestibility and Blood Lipids in Rats Fed a High Fat Diet

摘要


幾丁聚醣抑制腸道脂肪消化率之作用可能和其分子量大小有關,因此,本研究探討以酵素水解法製備之高分子量幾丁聚醣對餵飼高脂飼料之大白鼠體重、蛋白質與脂肪消化率及血脂質濃度之影響。50隻10周齡大之雄性Wistar大鼠依體重隨機分配成五組,均餵飼高脂飼料分別含5%甲基纖維(對照組)、5%幾丁聚醣(1200 KDa, A組;600KDa, B組;300KDa, c組;150KDa, D組)之半純化試驗飼料共四週。飼養期間每日紀錄老鼠體重及攝食量,並收集糞便以測定糞便內容物。在第28天犧牲老鼠取血液以進行血脂質分析,並秤量肝臟及副睪脂肪塊重。結果顯示,幾丁聚醣組之相對副睪脂肪塊重顯著的較對照組低,顯示餵飼幾丁聚醣可減少體脂肪囤積。添加幾丁聚醣可增加排便量、糞便含水量及中性固醇排泄量。A組及B組血清三酸甘油酯濃度顯著降低,然而血膽固醇濃度卻不受影響。餵飼幾丁聚醣亦顯著的降低膳食脂肪粗消化率,且隨著幾丁聚醣分子量增加而有下降趨勢,然而,餵飼分子量1200KDa幾丁聚醣並無法進一步降低脂肪粗消化率。以上結果顯示,在攝取高脂飼料情況下,高分子量幾丁聚醣可經由抑制攝食量及膳食脂肪消化,有效的降低大白鼠增重和體脂肪的囤積。分子量三1200KDa之幾丁聚醣可能會因在胃中不易溶解而影響其抑制脂肪消化作用。

The inhibitory effect of chitosan on dietary fat digestion is partly related to its molecular weight (Mr). The purpose of this study was to investigate the effect of high-molecular-weight chitosans on nutrient digestibility, body weight gain and serum lipid profiles in rats. Fifty male, 10 wks old Wistar rats were randomly assigned into 5 experimental groups and fed a 15% high-fat diet with 5% cellulose (Control group), or 5% chitosans (A group, 1200 kDa Mr; B group, 600 kDa Mr; C group, 300 kDa Mr; D group, 150 kDa Mr) for 4 wks. Body weight and feed intake were recorded daily. Feces were collected for the analysis of fecal contents. At the end of feeding period, blood samples were collected and epididymal fat pad weight was measured. The results showed that relative epididymal fat pad weight was decreased in all chitosan groups indicating a reduced accumulation of body fat by chitosan feeding. When compared to the control group, fecal excretion, moisture and neutral steroids contents were increased significantly in chitosan groups. Serum TG was decreased in A and B groups, however serum TC was not affected in rats. The apparent fat digestibility was decreased significantly by chitosan feeding, and tended to be decreased as Mr of chitosan increased, however, no difference on fat digestibility was observed between group A and group B. The results of this study suggest that high Mr chitosans are effective in reducing body fat by inhibiting feed intake and dietary fat digestion in rats fed on a high fat diet. However, chitosans with Mr of1200 kDa, probably due to a poor solubility, are less effective than 600 kDa chitosans in inhibiting dietary fat digestion.
