孵化時間對受精雞蛋蛋黃成分及其促進雞胚胎纖維母細胞增生能力之影響

Effect of Incubation Time on Fertilized Chicken Egg Yolk Composition and Their Ability of Promoting the Proliferation of Chicken Embryonic Fibroblast Cell

摘要


本研究在探討受精雞蛋蛋黃於0-5天孵化期之成分變化,及應用細胞培養模式評估其生物活性。結果顯示,受精卵蛋黃之蛋白質、脂肪、膽固醇、磷脂質及三酸甘油酯等含量均隨孵化天數增加而減少,水分含量則隨孵化天數而增加,灰分在孵化0-4天無明顯變化;IgY抗體含量在孵化三天達最高量 (29.67 mg/mL),而後下降,至孵化五天時降為13.39 mg/mL。在雞胚胎纖維(Chicken Embryonic Fibroblast; CEF)細胞培養試驗中,所有試驗蛋黃溶液樣品(孵化0-5天)均能刺激CEF增生,並以孵化三天之蛋黃溶液的效果最佳,且其最佳的添加量為細胞培養基(DMEM)的1%。本試驗的結果可提供進一步研製具生物活性產品之參考。

The objective of this study was to investigate the effect of hatch stages (0-5 days) on the composition of fertilized chicken egg yolk (FCEY) and assess its bioactivities by chicken embryonic fibroblast (CEF) cell culture model. Results showed the contents of protein, fat, cholesterol, phospholipids and triglycerides decreased and the moisture increased of FCEY with days of incubation increasing. Ash contents of FCEY didn't notably change in 4 days incubation period. IgY of that was the highest (29.67 mg/mL) at 3 days of incubation and then declined to 13.39 mg/mL at 5 days of incubation. On the CEF cell culture test, all of FCEY at hatch stages (0-5 days) could stimulate the proliferation of CEF cell, and the best of these was the FCEY at 3 days of incubation. Additionally, 1% FCEY (3 days of incubation) in DMEM medium was the optimum dosage for CEF cell culture test. The results were a novel message for furthering research and development of egg yolk products with bioactive function.
