不同顏色植物中的粗多醣對小鼠初代脾臟細胞分泌細胞激素之影響

Effects of Crude Polysaccharides from Plants with Different Colors on Cytokine Secretions Using Mouse Primary Splenocytes

摘要


傳統中草藥或天然蔬果分離出的多醣可能具有生物活性,本研究為比較不同顏色植物來源多醣之免疫調節活性,選用四種食材經熱水萃取,再以70%酒精沉澱分離出粗多醣,分別獲得蓮子心粗多醣(lotus plumule crude polysaccharide, LPPS)、黑木耳粗多醣(tree mushroom crude polysaccharide, TMPS)、白甘藍菜粗多醣(white cabbage crude polysaccharide, WCPS)及紫甘藍菜多醣(red cabbage crude polysaccharide, RCPS),取這四種粗多醣非細胞毒性劑量進行研究,探討單獨添加或對以脂多醣(lipopolysaccharide, LPS)刺激對脾臟細胞分泌細胞激素的影響。結果顯示,單獨添加LPPS與TMPS時,隨著添加濃度的增加,脾臟細胞促發炎的腫瘤壞死因子-α(tumor necrosis factor-α, TNF-α)/抗發炎的介白素-10(interleukin-l0, IL-10)細胞激素分泌比值隨之降低,顯示LPPS與TMPS均具有抗發炎潛力。在以LPS刺激的脾臟細胞發炎模式下,LPPS隨著添加濃度的增加,脾臟細胞TNF-α/IL-l0細胞激素分泌比值隨之降低。綜合本研究結果建議,四種不同顏色植物中的粗多醣,以蓮子心粗多醣最具抗發炎潛力,黑木耳粗多醣次之。

Polysaccharides from traditional Chinese herbal medicine, fruits or vegetables may have potent biological activities. To compare immunomodulatory activities of polysaccharides from plants with different colors, lotus plumule crude polysaccharides (LPPS), tree mushroom crude polysaccharide (TMPS), white cabbage crude polysaccharides (WCPS) and red cabbage crude polysaccharides (RCPS) were extracted with hot water and then precipitated using 70% alcohol. Non-cytotoxic doses of the crude polysaccharides were administered to mouse primary spleriocytes in the absence or presence of lipopolysaccharide (LPS) to investigate the effects of polysaccharides from plants with different colors on cytokine secretions. The results showed that LPPS and TMPS, respectively, treated in the absence of LPS, significantly decreased pro-/anti-inflammatory cytokine (TNF-α/IL-10) secretion ratios in dose-dependent manners, indicating that LPPS and TMPS have anti inflammatory potentials. Furthermore, LPPS treated in the presence of LPS significantly decreased TNF-α/IL-10 secretion ratios by splenocytes in a dose-dependent manner. Overall, our findings suggest that LPPS has the strongest anti-inflammatory effect among four crude polysaccharides from plants with different colors selected in this study. TMPS had a mild anti-inflammatory effect.
