氯化鉻對Streptozotocin及Nicotiamide誘導之糖尿病大鼠血糖與血脂的影響

Effect of Chromium Chloride on Plasma Glucose and Lipids in Diabetic Rats Induced by Streptozocin and Nicotinamide

摘要


為探討加三價鉻對血糖與血脂影響,以鏈佐黴素(streptozotocin;STZ)及菸鹼醯胺(Nicotinamide)誘發之第二型糖尿病大鼠為試驗動物,餵食加乳鉻粉。動物共分為六組,分別是(1)正常組(N);(2)糖尿病控制組(D);(3)0.5倍劑量乳鉻組(0.5D:4.2微克三氯化鉻);(4)一倍劑量乳鉻組(1D:8.3微克三氯化鉻);(5)三倍劑量乳鉻組(3D:25微克三氯化鉻);(6)0.8 g/kg BW rosiglitazone (TZD)組。試驗期間餵食高脂飼料,每週測量體重,試驗共進行九週。並於試驗第四週及第八週,進行葡萄糖耐受性測試(OGTT)。試驗結果發現,試驗四週後,糖尿病大鼠給予不同劑量之三價鉻粉OGTT與糖尿病控制組相比可顯著降低第30、60及120分鐘之血糖濃度。試驗八週後,給予糖尿病大鼠不同劑量之三價鉻粉均可以降低OGTT第60及180分鐘的血糖濃度。糖尿病大鼠給予0.5X及1X劑量三價鉻粉九週後空腹血漿葡萄糖顯著低於糖尿病控制組,但胰島素、HOMA-IR及QUICK index值方面,給予糖尿病大鼠不同劑量之三價鉻粉與糖尿病控制組無顯著差異。另一方面,一倍劑量乳鉻組明顯降低肝臟膽固醇及三酸甘油酯含量但不影響血漿膽固醇及三酸甘油酯濃度。在肝腎功能指標方面,三價鉻粉不會影響血漿creatinine值,但可顯著降低糖尿病大鼠血漿AST及ALT活性。綜合上述結果推測,在大鼠中,三價乳鉻粉有助於調節血糖。

To investigate the effect of chromium chloride on plasma glucose and lipids, Sprague Dawley (SD) normal rats and diabetic rats induced by streptozotocin and nicotinamide were used as experimental animals. Rats were divided into 6 groups, which were (1) normal group (N), (2) diabetic group (D), (3) diabetic with CrCl3 (4.2 μg; 0.5D) group, (4) diabetic with CrCl3 (8.3 μg; 1D) group, (5) diabetic with CrCl3 (25 μg; 3D) group, and (6) diabetic with TZD group. Diet and drinking water fed to rats were ad libitum. The experiment was done for 9 weeks. After 4 weeks of feeding, chromium chloride significantly decreased plasma glucose level at 30, 60, 120 min after glucose tolerance challenge, but only decreased plasma glucose level at 120 and 180 min after 8 wk treatment. Lower fasting plasma glucose levels were observed in 0.5D and 1D groups when compared with the control group. However, no significant difference in insulin level, HOMA-IR and QUICK index was found between the control group and the chromium chloride supplemented group. It is interesting to note that chromium chloride did not change plasma cholesterol and triglyceride levels, but decreased hepatic cholesterol and triglyceride contents. Although chromium chloride did not affect plasma creatine values, it reduced AST and ALT activities. It is suggested that chromium chloride supplementation can regulate glucose metabolism in diabetic rats.
