以糊化澱粉糊黏著劑探討造粒特性及其貯存安定性

Study on Gelatinized Starch Solution as Binder to Investigate the Granulation Property and Storage Stability

摘要


粉粒(powder)是食品、醫藥、化工產業常用之產品型態,流動床造粒常利用黏著劑、賦型劑(澱粉顆粒)進行濕式黏著、包覆造粒之目的。本實驗以羧甲基纖維素、玉米澱粉、樹薯澱粉糊化液為黏著劑探討造粒可行性及其粉粒品質、貯存安定性,實驗結果如下:以0.5% CMC, 63.67 cP為黏度指標,玉米澱粉糊化液2.78%、樹薯澱粉糊化液1.87%可達到相似黏度;三種黏著劑造粒之粒徑均隨造粒時間延長呈提高,造粒180 min可達到造粒最大值,其中以CMC粒徑最大(164.64 μm);造粒粉粒外觀(SEM)均呈小顆粒黏著形成大顆粒之外表,其色素(a值)包覆量三者之間沒有顯著差異(p > 0.05)。在粉粒品質分析中,以玉米澱粉糊化液造粒粉粒之體積密度、振實密度較低,流動性以CMC、樹薯澱粉糊化液造粒粉粒較低;三種粉粒在貯存期間均隨溫度、濕度提高,時間延長其水分、水活性均呈增加變化,其中以第一天增加較快,包覆量a值則變化不顯著。由本實驗結果澱粉糊化液可作為造粒之黏著劑用途,其粉粒品質與傳統CMC造粒相近。

Powder is a common product form in food, medicine and chemical industry. Fluid bed granulation often used binder and excipient (starch particle) for wetting adhesion and coating granulation. In this experiment, we used carboxymethyl cellulose, corn starch and tapioca starch gelatinized solution as binder to investigate the feasibility of granulation, quality of powder and its storage stability. The experimental results were as follow: using 0.5% CMC solution as a standard, 2.78% corn starch gelatinized solution and 1.78% of tapioca starch gelatinized solution can get similar viscosity. With granulation time increasing, the particle size of powder granulated with three kinds of binders was getting bigger. When the powder granulated for 180 min reached the biggest particle size, and the particle size of the powder granulated with CMC is the biggest among all (164.64 μm). Appearance of powder under SEM showed that bigger powder was formed by agglomeration of small particles. The encapsulated amount of the color (a value) showed no significant difference between three (p > 0.05). In the analysis of the power's quality, powder granulated with corn starch gelatinized solution showed lower bulk density and tapped density; powder granulated with CMC and tapioca starch gelatinized solution showed lower flowability. As temperature, humidity and time increasing, the water content and the water activity of the powder increased during storage. And it showed a more rapidly increasing in the first day. The color (a value) of the powder did not change significantly during storage. The research results showed that gelatinized starch solution can be used as binders for granulation process, its quality of the powder was similar with conventional granulation (CMC).
