Isolation and Identification of a Novel Feline Coronavirus from a Kitten with Naturally Occurring Feline Infectious Peritonitis in Taiwan

自貓傳染性腹膜炎自然感染病例中分離與鑑定台灣貓冠狀病毒

摘要


貓傳染性腹膜炎(Feline infectious peritonitis; FIP)為慢性、漸進性、致命性疾病,其致病原為貓冠狀病毒(Feline coronavirus; FCoV)。截至目前為止,有關此疾病致病機制之研究皆得自少數的病毒株,而較近的病毒株分離至今也已將近20年。本研究由臨床上一隻4.5月齡小貓自然發生之FIP病例,自其胸水與貓全胚胎細胞株進行共同培養(Co-cultivation),成功地分離到本土株貓冠狀病毒。共同培養後三天可觀察到冠狀病毒感染所引起的典型多核巨大細胞之細胞病變效應。經由免疫染色進行其抗原辨認並以聚合酵素鏈鎖反應進行病毒分型,確認此分離株為第二型FCoV。在病毒的生長曲線中(one-step viral growth curve),發現此分離株於感染細胞後6小時內即可完成一個病毒複製循環,複製所需的時間較其他國外的FCoV病毒株為短。目前已知的第二型貓冠狀病毒皆能於貓全胚胎細胞及貓腎細胞生長良好,但本分離株僅能在貓全胚胎細胞上快速生長。綜合以上結果發現這是一個生長特性異於現存FCoV的新病毒分離株,為台灣的首度報告,其命名為FCoV/NTU156/P/07(NTU156)

Feline infectious peritonitis (FIP) is a chronically progressive and usually fatal disease caused by feline coronavirus (FCoV). Only few virus strains were documented around the world; the latest FCoV strain was an isolate obtained nearly twenty years ago. By co-cultivation of pleural effusion from a 4.5-month-old FIP kitten with feline fcwf-4 cells, a local FCoV was isolated. Multinucleated cells, a typical coronavirus induced cytopathic effect (CPE), were readily observed after three days of co-cultivation. Viral antigen was confirmed by immunostaining and the virus was identified as a type II FCoV through genotyping. A replication cycle less than 6 hours, shorter than any other FCoV reference strains, was noted. Among the two feline cell lines that were reported to propagate type II FCoV, only fcwf-4 cells appear to support the growth of our isolate efficiently. This novel FCoV isolate with distinct growth characteristics is denominated as FCoV/NTU156/P/07 (NTU156), the first FCcV strain in Taiwan.
