臺灣香檬果皮成分分析、抗氧化及抑制一氧化氮活性之研究

Component Analysis and Antioxidant and Anti-nitric Oxide Activities of Peel of Citrus Depressa Hayata

摘要


臺灣香檬(Citrus depressa Hayata)果皮為研究材料,以nobiletin、tangeretin、hesperidin、diosmin及5-hydroxy-6,7,8,3’,4’-pentamethoxyflavone等成分作為指標,進行不同果徑大小果皮之High Performance Liquid Chromatography(HPLC)定量分析,並將果皮以methanol(MeOH)浸泡之萃取物,經分配層析離法(partition chromatography),分成ethyl acetate(EA)、n-butanol(n-BuOH)及水層可溶物,探討各分層之抗氧化及抑制nitric oxide (NO)生成的活性。結果顯示,各指標成分的含量皆隨果徑的增加而有下降的趨勢,僅diosmin於大果中具有較高的含量。各分層中以n-BuOH層萃取物具有較佳之自由基清除能力,其IC50為370 μg mL-1;此外,EA層可溶物具有較佳抑制NO生成的活性,並進一步利用管柱層析追蹤篩選其活性成分區段,結果顯示,noliletin及tangeretin成分含量較高的區段4、5,具有較佳抑制NO生成的活性,而各指標成分中以noliletin具有最佳的抑制活性。

The objective of this study was to determine the contents of nobiletin, tangeretin, hesperidin, diosmin, and 5-hydroxy-6,7,8,3’,4’-pentamethoxyflavone in the peel of different sizes of Hirami lemon (Citrus depressa Hayata) fruits by using High Performance Liquid Chromatography. The chemicals in the citrus peels were extracted with methanol, and then separated with ethyl acetate, n-butanol and water layer for partition chromatography. The antioxidant and anti-nitric oxide activities of each layer were also evaluated. Results indicated that the contents of marker components were decreased with the increasing of fruit size, except diosmin. Antioxidant and anti-nitric oxide activities tests showed that n-BuOH layer had the highest antioxidant activity and its IC50 was at 370 μg mL-1. However, the EA layer had the best anti-nitric oxide activity. The bioactive fractions were screened by column chromatography, showing that fractions 4 and 5 had higher contents of noliletin and tangeretin and better anti-nitric oxide activity. Nobiletin was the tested compound that expressed the best anti-nitric oxide activity.
