馬齒莧在不同粒徑下之抗炎效果研究

The Anti-inflammatory Effects of Portulaca Oleracea L. with Different Particle Sizes

摘要


本試驗採用96微米與5微米粒徑的馬齒莧(Portulaca oleracea L.)之溶液,依不同濃度分別作用於巨噬細胞株(mouse BALB/c macrophage RAW 264.7)進行抗發炎活性之檢測,包括一氧化氮(nitric oxide, NO)、白介素6(interleukin 6, IL-6)及腫瘤壞死因子-α(tumor necrosis factor-alpha, TNF-α)等三種細胞發炎激素,以及細胞毒性試驗(microculture tetrazolium assay, MTT assay)。結果顯示,96微米粒徑的馬齒莧,在5.0%及10.0 %之測試濃度下,均可顯著抑制IL-6與TNF-α的產生,而NO則無抑制效果。5微米粒徑的馬齒莧,在0.25%、0.5%及1.0 %之測試濃度下,皆可顯著抑制IL-6與TNF-α的產生,而在NO則無抑制效果。同時96及5微米粒徑的馬齒莧兩者在MTT細胞毒性檢測方面,皆無顯著之細胞毒害作用。結論,使用96微米與5微米粒徑的馬齒莧溶液,具顯著抑制IL-6與TNF-α生成之抗炎效果,且未具細胞毒性。此外,粒徑較小之5微米馬齒莧溶液,除有顯著抗炎效果外,更具節省材料與成本之潛能效益,提供馬齒莧進一步的研究與開發應用。

The purpose of this study was to evaluate the effects of 96-micron and 5-micron Portulaca oleracea L. on anti-inflammatory activity of cells. Three pro-inflammatory factors were detected on macrophage cell line (mouse BALB/c macrophage RAW 264.7), including nitric oxide (NO), interleukin 6 (IL-6) and tumor necrosis factor-alpha (TNF-α), and the cytotoxicity (microculture tetrazolium assay, MTT assay) was analyzed. The results showed that 5.0 %, 10% concentration of 96-micron Portulaca oleracea L. solutions administration on cell could inhibit the production of IL-6 and TNF-α, but NO were not affected. The 0.25%, 0.5%, 1.0% concentration of 5-micron Portulaca oleracea L. solutions administration on cell could inhibit the production of IL-6 and TNF-α, but NO were not affected. MTT assay showed that the solutions of 96-micron and 5-micron Portulaca oleracea L. administration had no significant effects on the survival rate of macrophages. The conclusion was that the 96-micron and 5-micron solutions of Portulaca oleracea L. could significantly inhibit the inflammatory effects of IL-6 and TNF-α, and no cytotoxicity. The 5-micron Portulaca oleracea L. solution not only has significant anti-inflammatory effect, but also saves materials and costs. The results could be provided a more in-depth basis for the research of Portulaca oleracea L.
