化粧品中染髮劑成分分析方法之建立

Determination of Hair Dye Ingredients in Cosmetics by Liquid Chromatography-Tandem Mass Spectrometry

摘要

本研究係利用高效液相層析串聯式質譜儀(LC/MS/MS)建立一簡便且快速的方法，檢體以50%甲醇水溶液溶解後，經ACQUITY UPLC HSS T3層析管柱(2.1×100 mm, 1.8 μm)，採用10 mM甲酸銨水溶液(以氨水調整pH值至9)及甲醇/乙腈(1:1, v/v)為移動相作梯度沖提，並以多重反應監測模式(MRM)偵測，可同時分析染髮劑中2,6-diaminopyridine、phenyl methyl pyrazolone、5-amino-o-cresol、m-phenylenediamine、2,4-diaminophenoxyethanol、2-nitro-1,4-diaminobenzone、m-aminophenol、N-phenyl-p-phenylenediamine、p-phenylenediamine、p-aminophenol、o-phenylenediamine、p-methylaminophenol、4-amino-3-nitrophenol、4-chlororesorcinol、1-naphthol、4-amino-m-cresol、o-aminophenol、toluene2,5-diamine、o-chloro-p-phenylenediamine、p-nitro-o-phenylenediamine、6-amino-mcresol、1,5-dihydroxnaphthalene、6-hydroxyindiole、2-methylresorcinol及resorcinol等25種成分。25種染髮劑成分之標準曲線線性範圍為0.005-250 mg/mL，標準曲線線性回歸係數R2皆為0.995以上。添加回收試驗結果，回收率介於78.5-116.7%，相對標準偏差均小於8.6%。應用本研究所建立之方法檢測市售染髮劑共10件，結果顯示應用本方法可適用於市售染髮劑產品之檢驗，本方法將提供各界檢驗之參考。

A specific and rapid liquid chromatography-tandem mass spectrometry (LCIMSIMS) method was developed to simultaneously determi ne 25 hair dye ingredient, including 2,6-diaminopyridine, phenyl methyl pyrazolone, 5-amino-o-cresol, m-phenylenediamine, 2,4-diaminophenoxyethanol, 2-nitro-l,4diaminobenzene, m-aminophenol, n-phenyl-p-phe nylenediamine, p-phenylenediamine, p-aminophenol, o-phenylenediamine,p-methylaminophenol, 4-amino-3-nitrophenol, 4-chlororesorcinol, I-na phthol, 4-amino-m-cresol, o-aminophenol, toluene 2,5-diamine, o-chloro-p-phenylenediamine, p-nitro-ophenylenediamine, 6-amino-m-cresol, 1,5-dihydroxynaphthalene, 6-hydroxyindiole, 2-methylresorcinol and resorcinol in commercial hair dye products. The linearities and recoveries of the method were observed at the concentration range of 0.005-250 ug/ml. and 78.5-116.7%, respectively. The method had been successfully applied to screen 10 commercial hair dye products. The samples were prepared in 50% methanol and then separated using an Acquity UPLC HSS T3 (In 2.1 mm x 100 mm, 1.8 urn) column with gradient elution containing 10 mM ammonium formate (adjusted to pH 9 with ammonia) and methanol/acetonitrile (1:1, v/v), The survey indicated that all the samples complied with the regulatory requirments.
