中部地區彩色海芋病毒病害發生調查及一種Tospovirus之分離

Field Survey of Virus Disease and Characterization of a New Tospovirus on Calla Lily in Central Taiwan

摘要


利用indirect ELISA分析法,以CMV、TuMV、DsMV及ZaMV等4種植物病毒抗血清調查中部地區11個彩色海芋園之病毒發生狀況。調查結果顯示,不同彩色海芋園病毒罹病程度為2.6~70.8%不等。總計採集261個擬似病毒罹病植株中,indirect ELISA檢測結果有191個樣品產生正反應,其中單獨感染1種病毒者佔53.4%,複合感染二種以上病毒者佔46.6%。單獨感染病毒之102株彩色海芋中,感染ZaMV佔50.0%、DsMV佔23.5%,TuMV佔19.6%及CMV佔6.9%。複合感染病毒之89株彩色海芋中,複合感染二種病毒者佔83.1%、複合感染三種病毒者佔14.6%、複合感染四種病毒者佔2.3%。自彰化縣北斗鎮栽培之彩色海芋分離到1種新tospovirus,稱之彩色海芋黃斑病毒(CCSV)。罹病植株於中位葉片出現淡綠至黃化斑。陰染罹病葉粗汁液或病組織超薄切片,電子顯微鏡可觀查到75~105 nm類似球形之病毒顆粒。以病組織粗汁液機械接種36種植物,其中包括冬瓜及矮南瓜等22種植物對CCSV病毒呈感受性。CCSV可經由南黃薊馬傳播。Indirect ELISA顯示CCSV與WSMoV具有血清類緣關係,但與INSV、TSWV、PCFV則無。

During the survey of the virus infection in the calla lilies gardens in central Taiwan, eleven gardens were randomly sampled and analyzed by indirect ELISA with CMV, TuMV, DsMV and ZaMV antisera. The results indicated the virus incidence of calla lilies varied from the gardens. Of the total 191 from 261 virus-infected plants, 53.4% of infection are being infected by one virus, among these, 50.0% were infected by ZaMV; 23.5% were infected by DsMV; 19.6% were infected by TuMV and 6.9% were infected by CMV. The rest 46.6% of infected plants were complex infection. Among these, 83.1% were infected by two viruses, 14.6% were infected by three viruses and 2.3% were infected by four viruses. During the survey, a new tospovirus, designated as Calla lily chlorotic spot virus (CCSV), was isolated from calla lilies in Changhua county. Chlorotic spots appear on middle leaves of the infected plants. Spherical virions of 75-105 nm in diameter, similar to that of tospovirus presented in crude extracts and ultrathin sections of diseased leaves. Among 36 plant species inoculated mechanically, 22 species of plants including wax gourd and zucchini squash were susceptible to the virus. CCSV was transmitable by Thrips palmi to healthy wax gourd and zucchini squash. Serological tests (indirect ELISA) showed that the CCSV antigen did react with antisera CCSV and WSMoV, however,did not react with that serum against TSWV, INSV, and PCFV.
