Capillary Electrophoresis for Reaction Optimization of Combinatorial Synthesis

Combinatorial chemistry approaches have facilitated the research and development process of lead discovery and optimization. A successful combinatorial program typically includes high throughput library synthesis , characterization, and screening, and lead identification and confirmation. One of many challenges in this program is to develop high throughput characterization methods to address issues concerning reaction stereoselectivity and possible racemization during the library synthesis. Capillary electrophoresis is a high resolution separation technique, which is well suited for the stereoisomer characterization of libraries of small organic molecules. In this report, we demonstrate the usefulness of capillary electrophoresis in optimizing reactions, determining stereoisomer ratios, and assessing reaction racemization during combinatorial synthesis.
