摘要


本文介紹2,5-dimethoxy-4-ethylthiophenethylamine (2C-T-2)對老鼠的脾臟及胸腺淋巴細胞內NO活性的影響。研究發現當2C-T-2餵食後,小鼠脾臟的T-和B-淋巴細胞與胸腺的T-淋巴細胞經過活化後,細胞產生NO的量都有明顯的減少。這代表2C-T-2會干擾免疫細胞活性,因為NO的減少表示免疫能力的降低。另一方面,實驗以腹腔皮下注射方式對三隻大鼠給藥(20mg/kg),並收集注射前後24小時的尿液,運用GC-SIM-MS 的方法進行2C-T-2在老鼠體內的代謝路徑的分析。尿液樣品經過加酸加熱水解後,以乙醚液-液萃取方式萃取代謝物。實驗結果發現四種主要可能的代謝物,並由此推測2C-T-2在老鼠體內的代謝路徑。

The bioactivity of 2,5-dimethoxy-4-ethylthiophenethylamine (2C-T-2) on nitric oxide (NO) production and the proliferation of spleen and thymus lymphocytes to mitogen stimulation in mice are reported. NO production by T and B lymphocytes in spleen and T cells in the thymus of mice decreased after the oral administration of 2C-T-2. This indicates that 2C-T-2 intake may perturb both neural and immune activity since a decrease in NO production is indicative of a weakened defense function. On the other hand, a simple and specific method based on gas-chromatography-selected ion monitoring mass spectrometry (GC-SIM-MS) for the analysis of in vivo metabolism of 2C-T-2 in rats is described. Three male rats were administered 20 mg/kg of 2C-T-2 by intra-peritoneal injection, and 24 h urine fractions were collected before and after the administration for analysis. After acidic hydrolysis of the urine samples, the metabolites were liquid-liquid extraction and analyzed by a quadruple mass spectrometer in the selected ion monitoring mode. The findings show that four metabolites are present and the metabolic pathway for 2C-T-2 in the rat is proposed.
