The Colchicine Derived from Ethanol Extract of Daylily Hemerocallis fulva L. Flower in Taiwan

台灣金針花乙醇萃取物含秋水仙素衍生物

摘要


Colchicine is one of very soluble chemical in ethanol that well noted in books. The ethanol extract from the freeze dried flower powder of daylily Hemerocallis fulva L. was separated through thin layer chromatography (TLC) for screen test at first. There was one spot in testing material and the Rf value was as similar as the standard of colchicine. The spot was scratched down and re-extracted by ethanol, then analyzed by U V spectrophotometer and high performance liquid chromatography (HPLC). The retention times of colchicine standard and ethanol extract tested sample were 12.517 and 12.213 min, respectively. The UV scanning and LC/MS/MS comforming showed the different types of diagram, MW peak and major daughter peak. The main fragments of colchicine standard have m/z at 400.4 and 310.2, but the unknown compound has m/z at 444.2, 400.4 and 258.4. The result showed that there is no cochicine contianing in the daylily's flower which grow at Tai-Dong Mountain in Taiwan. The diarrhea of daylily flower may be outcome from colchicine derived,that were another alkaloids or steroids.

因秋水仙素對乙醇有極高的溶解度,故將金針花乾燥粉末以乙醇萃取,其萃取物經薄層層析(TLC)法,作秋水仙素初步篩選試驗,發現金針花抽出物在與秋水仙素標準品相同Rf值處具有相似之斑點,將此斑點刮下以乙醇再萃取,並以高效液相層析儀及紫外線分光光度計進行分析。結果發現秋水仙素標準品之滯留時間(12.517 min)與金針花乙醇萃取物滯留時間(12.213 min)雖差異不大,然再經LC/MS/MS及UV 掃瞄,顯示不僅兩者之圖譜不同,其M.W. peak及Daughter peak亦不一,標準品之m/z在400.4及310.2,而樣品乙醇萃取物則在258.4, 400.4及444.2。由結果知台灣台東地區之金針花不含秋水仙素,此結果與民問傳述造成腹瀉的原因不儘相同,有待更深入研究。
