Moringa oleifera, Lam Seeds Extract as Anti Atherosclerosis Through Decreasing of LDL Level and the Intimal-Medial Thickness of Aorta in High- Fat High-Fructose Fed Rats

Introduction: High-fat high-fructose diets are strongly correlated with the formation of atherosclerosis plaque that cause an increase in stroke and coronary heart disease due to raise of LDL level and oxidative stress on endothelial wall. Moringa seeds (Moringa oleifera Lam.) contains secondary metabolites such as β-sitosterol and antioxiodants that can prevent atherosclerosis. The aim of this study is to investigate the effect of ethanolic extract of Moringa seeds on Low Density Lipoprotein level and Intimal-Medial thickness of aortic wall in High-Fat High-Fructose Fed rats (Rattus norvegicus) as anti atherosclerosis. Methods: This laboratory experimental method study design used twenty-eight male rats which were assigned into 4 groups. K1 (control) were fed with standard pellet, meanwhile K2, K3 and K4 were fed with high-fat high-fructose diet for 50 days then LDL level is measured. Ethanolic extract of Moringa seeds were administered at dose of 150mg/kg body weight for K3 and 200mg/kg body weight for K4. After 28 days, LDL levels were measured, rats were terminated and thoracic part of aorta were taken. LDL levels were analysed using Paired T Test and One Way ANOVA test. Intimal-media thickness was measured aortic wall cross section then analysed using Kruskal Wallis test and Mann Whitney as the post hoc. Result: Paired T Test showed a significant difference between the mean of LDL level before-after high-fat high-fructose diet and after Moringa seeds extract treatment (p < 0.05). One-Way ANOVA test and Tukey post-hoc test showed significant difference between K1-K2, K1-K3, K1-K4, K2-K3, and K3-K4. Meanwhile Kruskal-Wallis and Mann-Whitney post hoc test showed significant differences in intimal-media thickness of aortic wall between K1-K2, K2-K3, K2- K4 group (p<0.05). Conclusions: Moringa oleifera seeds extract at dose of 150 mg/kgBW and dose of 200 mg/kgBW significantly reduces LDL level and Intimal-Media Thickness of Aortic wall in high-fat high-fructose fed rats (Rattus norvegicus).
