臺灣扁柏葉子熱水抽出物之抗氧化活性及延緩老化效應

Antioxidant Activity and Delayed Aging Effects of Hot Water Extract from Chamaecyparis obtusa var. formosana Leaves

摘要


本研究係探討臺灣扁柏 (Chamaecyparis obtusa var. formosana) 葉子熱水抽出物之抗氧化及延緩老化活性。首先以清除DPPH自由基活性、總酚類含量及細胞存活率比較不同萃取條件對臺灣扁柏葉子抽出物抗氧化活性及細胞毒性之影響,所獲得之最佳條件為以棲蘭山地區臺灣扁柏葉子熱水萃取6 hr,其抽出物抗氧化效果較佳且無細胞毒性。後續透過液相層析分離出臺灣扁柏葉子熱水抽出物之各可溶部,其中,乙酸乙酯可溶部總酚類化合物含量最高,故其清除DPPH自由基、清除超氧自由基及螯合亞鐵離子之能力最佳。而線蟲模式評估結果亦顯示,臺灣扁柏葉子熱水抽出物乙酸乙酯可溶部之抗氧化能力最優異,並可減緩老化蛋白之生成、減緩阿茲海默症狀及延長線蟲之壽命期。進一步以薄層層析、管柱層析及高效能液相層析分離乙酸乙酯可溶部之有效成分,並用質譜儀及核磁共振儀鑑定出6個化合物,分別為Catechin、Quercetin、Quercetin-3-O-α-rhamnpyranoside、Myricetin-3-O-α-rhamnpyranoside、Vanillic acid及 4-Hydroxybenzoic acid。本研究結果顯示臺灣扁柏葉子熱水抽出物極具潛力開發為抗氧化及延緩老化之保健食品,非常值得更深入研究。

The aim of this study was to investigate the antioxidant activity and delayed aging effect of hot water extracts from Chamaecyparis obtusa var. formosana leaf. First we evaluated the effect of different extraction conditions on the antioxidant activities of extracts from C. obtusa var. formosana leaf via the DPPH free radical scavenging assay, total phenolic contents test and cell viability test. Results showed that using hot water to extract the leaves of C. obtusa var. formosana collected from Mt. Chilan for 6 hr, the extracts with the best antioxidant activity and less cell toxicity were obtained. Liquid-liquid partition was employed to separate the hot water extracts of C. obtusa var. formosana leaves. Among all soluble fractions, the EtOAc fraction has the highest phenolic content, indicating that it has the best inhibitory effects on DPPH and superoxide free radicals and the best ferrous ion chelating activity. In the C. elegans model, the EtOAc fraction showed high antioxidant activity. Moreover, it decreased the deposition of lipofuscin, delayed β-Amyloid-induced paralysis and extended the lifespan. In addition, we also separated and identified constituents in the EtOAc fraction of C. obtusa var. formosana leaves by using thin layer chromatography (TLC), column chromatography (CC), high performance liquid chromatography (HPLC), mass spectroscopy and nucleus magnetic resonance (NMR). Six compounds were identified included catechin, quercetin, quercetin-3-O-α-rhamnpyranoside, myricetin-3-O-α-rhamnpyranoside, vanillic acid and 4-hydroxybenzoic acid. The results obtained revealed that the hot water extracts from C. obtusa var. formosana leaf has the potential to be used as a source for antioxidant or delayed aging health food and warrants further investigations.
