水稻與番茄種子活勢檢測探討

The study of several seed vigor tests in rice and tomato seed lots

摘要


種子活勢由多項特質組成,如種子的發芽速度、整齊度、健康幼苗比例以及在不適環境下的發芽能力,對於種苗生產來說是個至關重要的因子,但目前種子檢查多使用,在穩定的實驗室發芽環境進行的發芽率做為發芽表現的依據,往往無法吻合種子在環境不恆定的田間表現,本試驗旨在以水稻(Oryza sativa L.)與番茄(Solanum lycopersicum)種子建立完整的活勢檢測並與田間出土表現做相關性分析,檢測方法包含生理法與生化法,再以相關性分析選出吻合田間出土表現的活勢檢測方法。參試水稻種子為17批次發芽表現不一的種子,休眠程度也不一致,田間出土率介於2-100%;番茄種子13批次,田間出土率介於17-99%,因其中3批種子受到較低溫度的影響導致發芽延遲,因此進行相關性分析時僅選取10批種子。生理法包括加速老化法與胚根突出法,生化法包含TTC染色法與電導度法,實驗室紙上法的發芽參數及田間出土表現參數也列入相關性分析。水稻於45oC老化72h及番茄於41oC老化72h與FEP(Field final emergence percentage)取得高相關性,相關係數分別為0.928與0.937。水稻在23oC/168 h取得的胚根突出率與FEP取得最高相關係數0.951;番茄則是20oC/102 h取得最高相關係數0.986。番茄胚根突出法除了與FEP取得高相關性外,與MET(Mean emergence time)也具高相關性。水稻與番茄的生化法結果與田間表現的相關性都不高,兩作物之TTC (Triphenyl tetrazolium chloride)染色法與田間表現僅具中度相關,而電導度法與田間表現皆不具相關性。加速老化試驗耗時17天,TTC染色法為2天;水稻與番茄胚根突出法耗時不同,分別為168 h與78 h。綜合以上分析,胚根突出法省時、操作簡單且與田間出土速度、最終出土率都有高相關性,為最推薦的水稻與番茄種子活勢測定法。

Seed vigor is comprised of various characteristics such as germination speed, germination uniformity, normal seedling percentage, and germination performance under unfavorable environment. The germination obtained in a laboratory does not often match with field emergence, and both tests are time-consuming. The current study was aimed to establish vigor test methods for predicting field emergence in Rice (Oryza sativa L.) and tomato (Solanum lycopersicum) and the vigor test methods included physiological and biochemical methods. The seventeen tested rice seed lots have different germination and dormancy characters and the field emergence percentage range from 2-100%. The field emergence percentage of thirteen tomato seed lots range from 17-99% and three seed lots were affected by low temperature, resulting in lag germination, therefore, only ten tomato seed lots were used on correlation analysis. The physiological method includes accelerated aging method and radical emergence method and the biochemical method includes TTC dying method and electrical conductivity method were all tested against laboratory germination and field emergence parameters. The best accelerated aging condition for rice was aging at 45oC under high humidity for 3 days, and tomato was 41oC under high humidity for 3 days and the correlation coefficient against FEP was 0.928 and 0.937. The best condition for the rice and tomato seed radical emergence method was 23oC/168 h and 20oC/102 h and the correlation coefficient against FEP was 0.951 and 0.986, respectively. The tomato radicle emergence results also had a high correlation coefficient with MET. The biochemical method of both rice and tomato seed had a low correlation coefficient with FEP. The TTC dying test of rice and tomato seed lots has only a medium correlation coefficient with FEP and the electrical conductivity results of both seed lots were not correlated with field emergence parameters. The time cost for the accelerated aging method was 17 days, TTC dying was 2 days, rice seed radical emergence method was 168 h and tomato seed was 78 h. To sum up, the radicle emergence test is time-saving, conducted easily, and has a high correlation with both FEP and MET, is the best vigor test for rice and tomato seed.
