探討不同微/奈米結構之陽極氧化鋁模板運用於細胞培養之研究

摘要


本研究以比較微米、奈米之不同尺寸結構對細胞培養之影響,在實驗中製備四種不同結構(無結構、微米結構、奈米結構、雙重結構)之鋁片進行類骨母細胞(osteoblast-like cell, MG 63)拓撲反應之研究。 實驗中製作出三種不同尺度之氧化鋁薄膜,分別運用噴砂技術製作微米孔洞結構之氧化鋁薄膜;陽極處理製作一般奈米孔洞之陽極氧化氧化鋁薄膜;最後將具有微米孔洞之鋁模板進行陽極處理後,製作出在微米凹槽上擁有奈米孔洞之雙重結構。爾後將此不同結構之氧化鋁薄膜進行接觸角量測,探討成品表面特性(親水性、疏水性),同時研究製程參數與表面改質之關係。 最後將具氧化鋁薄膜於無塵室做滅菌處理,再以類骨母細胞培養於薄膜上,以MTT法,討論無結構、微米結構、奈米結構、雙重結構之差異,且對細胞在貼附、伸展、分裂繁殖之影響。

In this paper, we are investigating the different cultured structures scale in micrometer and nanometer. The experiment have been fabricated four types aluminum substrate structures which included none-structure、 micro-structure、 nano-structure、 and combined micro/nano structure to fulfill the research in osteoblast-like cell (MG 63). The experiment we have been made three scale thin film structures of Aluminum oxide, the micro-structure has been manufactured by sandblasting of an aluminum sheet, the nano-structure has been manufactured by anodized. Finally, the combined micro/nano structure has been manufactured by sandblasting of an aluminum sheet and anodization. Therefore, the experiment purpose are concentrating to measure the contact angle、 the surface properties (hydrophobia, hydrophile) and the optimized processing parameters in three types different scale Aluminum oxide thin film. Finally we performed the sterilization method to deal with the patterns of alumina sheet product in the clean room. Meanwhile, we demonstrated the osteoblast-like cell (MG63) to culture on the thin film. Finally, we have been used MTT assay study the behavior of cultured osteoblast-like cell to obtain the different properties between none-structure、micro-structure、 nano-structure and micro/nano structure for adhesion、 elongation and proliferation.
