血液凝結特性快速檢測之微流道晶片技術研發

摘要


血液中的抗凝血劑量檢測對於臨床手術來說是重要且必須的,其含量太多會造成術後血流不止,太少則會導致血液凝固於傷口而造成手術時的不便。近年來醫院在血液凝固的檢測上,大多使用PT及APTT的項目來測試,為了改善其在臨床量測的缺點,本團隊選擇使用電性及物理方式來觀測血液凝固時的變化,在電性的量測上,使用本團隊設計的電極來量測,利用血液凝固時所產生的阻抗變化來得知凝血時間,進而了解到血液中抗凝血劑的含量是否足夠;在物理方式的量測上,則是使用自製的微流道晶片來感測,利用流道的毛細現象來觀測血液流速,並依血液的流速來判斷凝血劑的劑量是否充足。結果顯示使用的方式可有效的分辨抗凝劑的多寡,在未來可實際應用於臨床的檢測上。

The detection of anticoagulant dosage in blood is important and necessary in clinical operations. With high anticoagulant dosage, it will cause heavy bleeding during surgery. In the opposite, it will lead to clotting problem with low anticoagulant dosage. To monitor the dosage, PT and APTT are used in hospitals. However, both PT and APTT are complicated and time-consuming. To overcome these obstacles, in this work, electrical impedance and physical viscosity are employed to monitor the anticoagulant dosage. For the electro-impedance analysis, different electrode design were designed and implemented to obtain the best electrode configuration for blood coagulation measurement. For the physical viscosity analysis, capillary microchannels with different channel width are used to monitor the viscosity characteristics of blood. By using both electro-impedance and physical viscosity methods, the dosage of anticoagulant can be distinguished. This work demonstrates a potential micro devices for anticoagulant dosage monitoring and it could be contributed in clinical applications.
