網版印刷碳電極生物感測器開發

Developing a biosensor base on screen printed carbon electrode

摘要


免疫球蛋白E(IgE)在近年來被認為是一種過敏反應的生物指標物,它在過敏者血液中的濃度和過敏反應嚴重程度息息相關。在傳統的IgE分析主要利用酵素免疫分析(ELISA),一般以酵素免疫分析為基礎下配合競爭法或者是常見的三明治法來進行抗原(antigen)和抗體(antibody)的辨識和分析,但在此情況下必須在抗原上標示螢光或使用較多的抗體、酵素才能進行定性和定量的相關分析;也因為需要使用較多的抗體,在測定上需要相當多的步驟和經費才能完成一個樣品的測定。 本研究利用活化網版印刷碳電極(Screen-Printed CarbonElectrode;SPCE),配合電化學聚合修飾方法聚合三聚氰胺(melamine)於SPCE表面,藉由poly(melamien)本身帶有的NH2,和交聯劑Glutaraldehyde(GA)反應達到固定抗體。形成poly(melamine)-Antibody(pME-Ab)的生物感測探針(biosensor probes)。經過固定的抗體再配合電化學方法Chronoamperometry(CA)進行抗原偵測,由於蛋白質為不良導體,在探針與抗原結合後利用由電化學的訊號改變程度來進行抗原的定量;探討網版印刷碳電極各種活化及探針修飾方式,並應用於偵測IgE,探討其線性範圍8-67 fM,偵測極限為2.7 fM。

Recently, the IgE (Immunoglobulin E) in the serum of a hypersensitivity patient is a useful index for the detection of allergy diseases. A common method for meassuring detection of IgE in serum is enzyme-linked immunoassay (ELISA) . This method requires many antibodies and is complicated. In addition, the materials and instruments of ELISA cost much. Electrochemical biosensors have reached more interests due to the advantages of good sensitivity, low cost, low reagent consumption and rapid analysis time. In this study, we used low-cost screen-printed carbon eectrode(SPCE) and electrochemical polymerization of melamine on SPCE surface to create one poly(mealmine) layer, then used glutaraldehyde(GA) as a cross-linked reagent to imobilize anti-IgE on SPCE. After immobilizing the anti-IgE, chronoamperometry(CA) analysis was implemented to examine the concentration of the target IgE. The linear range of IgE concentration was 8∼67 fM, and the detection limit was 2.7 fM.
