光學映射應用在小鼠心肌膜電位偵測與分析之研究

摘要


近期文獻中提出自動分析光學影像映射訊號的方法,可提供研究心臟電生理的科學家精確的演算數據資料並透過可視化方式將結果呈現。光學影像映射系統 (Optical Mapping System, OMS) 可有效地擷取心肌膜電位的光學訊號,但非正常型的膜電位活性 (Uneven Membrane Electrical Activity, UMEA) 目前仍需要手動檢測且不易判斷。本研究是透過區域最大最小值 (Local Maxima/Minima) 與模板匹配 (Template Matching) 的方法研究發展一自動偵測心肌膜電位偵測與分析的演算法。透過正常的小鼠心臟與具有酒精性心肌病變 (Alcoholic Cardiomyopathy)的小鼠心臟的病理資料進行本研發軟體分析驗證。並以乙型神經受體作用劑 (Isoproterenol, 0.5μM) 體外心臟直接灌流,並以光學影像映射系統記錄,以本研發軟體分析其觀測結果。從病理資料分析結果可知具酒精性心肌病的小鼠心臟上非正常型的膜電位活性較多,平均起源點約上升212.76個百分比。藥物實驗結果加入乙型神經作用劑後的平均起源點約上升8.71個百分比。並分別將分析結果進行 T 檢定,病理資料起源點分析結果進行 T檢定之 P 值為0.0029,藥物實驗起源點分析結果進行 T檢定之 P 值為0.003。其結果證明本實驗的演算法可用於非正常型的膜電位活性的偵測。本研究的結論為:本程式應用病理及藥理引起的非正常型的膜電位活性,實驗組數量明顯的發現增加,於光學影像映射系統可以被用來擷取心肌膜電位的光學訊號,透過自動圖像分析程式可分析心肌膜電位的光學訊號並大大提高精度和效率。

From literature review, it would seem that optical mapping is able to provide accurate analysis on cardioelectrophysiology and arrhythmia. Although optical mapping systems (OMS) are useful to acquire action potential effectively, uneven membrane electrical activity (UMEA) , which indicates a close correlation with leading to arrhythmia, cannot be detected by conventional OMS directly and still has to be verified by manual detection and verification with unavailable human manipulation errors and subject interpretation. Thus, the main purpose of the research is aimed to develop an algorithm to detect and analyze action potential automatically through comparison between signal local maxima/minima and template matching principles. The research has successfully developed new analysis software to compare pathology data between normal mice and murine hearts with alcoholic cardiomyopathy. Isoproterenol (0.5 μM) was infused into heart for experimental confirmation and an optical mapping system was also employed to record for action potential signals. From data analysis, it was found that the UMEA quantity of murine heart was increased with alcoholic cardiomyopathy. Compared with a reference setup the average of initial point of murine heart suffered with alcoholic cardiomyopathy was raised by 213.75%. Furthermore, by adding Isoproterenol, the average of initial point for medicine experiment’s result was raised by 8.71%, in which the level was increased from 1% to 9.71% if compared with the reference setup. Meanwhile, a T-test was run to verify results of medicine experiment and pathology data. The P value of pathology data was 0.0029 while the P value of medicine experiment was 0.003. This indicates that the developed method is capable of detecting UMEA. Moreover, the research also indicates that UMEA can be generated during repolarization. In pathologic and pharmacologic examinations, the UMEA was significant with high occurrence in the experiment group’s hearts that it could be detectable and analysis by the developed algorithm. From this, it indicates that the algorithm is able to detect and analyze the action potential of murine hearts by using the developed OMS, in which improved accuracy of action potential detection can be achieved for cardioelectrophysiology.
