不同界面上蛋白質對濃度及背景溶液之吸附行為探討

Effects of protein concentrations, bulk solutions and interfaces on protein adsorption

摘要


本研究以石英晶體微天秤作為感測重量變化的檢測器,探討裸金界面上蛋白質的吸附行為,選用牛血清蛋白(BSA)和溶菌酶(Lysozyme)作為模型蛋白,濃度範圍為0.01~1 mM。觀察背景溶液對蛋白質吸附行為的影響;結果顯示背景溶液中的鹽類濃度提高,會減少溶質與溶劑間的排斥作用力,提高蛋白質的吸附量。此外,溶菌酶在不同濃度下,對裸金界面具不同的吸附行為,低濃度的溶菌酶呈現多層吸附;高濃度則呈現單層吸附,推斷濃度差異造成鬆弛時間不同,表面所形成的蛋白質層厚度會影響後續蛋白質的吸附行為,根據此特性探討溶菌酶的吸附動力學。

This research investigated the adsorptive behavior of the protein on the chip of bare gold interface to signal response of mass sensor, quartz crystal microbalance(QCM).Use Bovine serum albumin (BSA) and lysozyme as the model of protein, range of concentration is 0.01 ~ 1 mM . Detecting the effect of the adsorption behavior of protein with different background solution; the results show that increasing in the concentration of salt in the background solution will reduce interaction of exclusion between solute-solvent, the amount of protein adsorption will reduce. When lysozyme has different adsorption behaviors on bare gold with different concentrations, lysozyme has multilayer adsorption in the low concentration; and has monolayer adsorption in the high concentration, the difference of relaxation time probably caused by the concentration, the thickness of protein layer on the surface will affect the subsequent adsorption behavior of protein, according to this characteristic to discuss the adsorption kinetics of lysozyme. When lysozyme has different adsorption behaviors on bare gold with different concentrations, lysozyme has multilayer adsorption in the low concentration; and has monolayer adsorption in the high concentrations, the different of relaxation time probably caused by the concentration, the thickness of protein layer on the surface will affect the subsequent adsorption behavior of protein.
