暴露於含有多環芳香碳水化合物的工作者其尿液中1-Hydroxypyrene濃度之相關性研究-以廚師與煉焦工人為例

摘要


本研究的目的是藉由利用生物偵測的結果(1-OHP、8-OHdG)瞭解女性廚師一工作週其暴露於油煙中1-OHP的變化情形,與評估1-OHP和8-OHdG的相關性,以及煉焦工人尿中1-hydroxypyrene(1-OHP)濃度的測定,以評估其暴露環芳香族碳氫化合物(Polycyclic Aromic Hydrocarbons,PAHs)的情形。 尿中1-OHP之測定方法是使用高效能液相層析儀附螢光偵測器進行分析。尿液中8-Hydroxy-2’-deoxyguanosine(8-OHdG)分析方法高效能液相層析儀串聯質普儀進行分析。 研究結果顯示,連續七天尿液中1-OHP與8-OHdG濃度分佈情形皆呈現多峰分佈;1-OHP與8-OHdG兩種指標濃度,並利用Spearman等級相關檢定法作相關性分析在一定的顯著水準,其對應的等級具有顯著的正相關性(R2=0.0589, p=0.0225)。 女性廚師尿中1-OHP的上下午之濃度分別為1.84及3.36(μmole/mole cr.)是對照組的1.1及3.2倍,對照組之尿中1-OHP濃度為1.66及1.07(μmole/mole cr.)。 煉焦爐工人(暴露組勞工)尿中1-OHP的濃度為44.2μmol/mol Cr.是非煉焦工人(對照組)的71.3倍,達統計上之顯著差異(P<0.05),非煉焦工人(對照組)之尿中1-OHP濃度為0.62μmol/mol Cr.。在本組勞工中,將抽菸者與非抽菸者分開來看,的確抽菸者中之煉焦爐工人(暴露組勞工)與非煉焦工人(對照組)的尿中1-OHP(53.0μmol/mol Cr. 與0.59μmol/mol Cr.)高過於非抽菸者中之煉焦爐工人(暴露組勞工)與非煉焦工人(對照組)的尿中1-OHP(38.4μmol/mol Cr. 與0.56μmol/mol Cr.),但皆不影響到暴露組與對照組之間尿中1-OHP濃度在統計上之顯著差異(P值皆<0.05)。 雖然女性廚師尿中1-OHP與對照組並無顯著性差異且低於煉焦爐工人,但其尿中1-OHP的上下午之濃度卻高達1.84及3.36(μmole/mole cr.),因此廚師有可能暴露於高濃度的PAHs,目前還未看到國內對於針對廚師油煙與PAHs生物指標1-OHP等相關研究,本研究結果可提供各界對職業安全衛生管理之參考。

The objective of this thesis was to study urinary excretion of 1-hydroxypyrene(1-OHP), a metabolite of pyrene, among workers (professional chefs and coke-oven workers) , who are potentially exposed to polycyclic aromic hydrocarbons(PAHs). In addition, the relationship between 8-hydroxy-2’-deoxyguanosine(8-OHdG), representative for oxidative stress, and 1-OHP in urine in coke-oven workers was examined . Urinary 1-OHP and 8-OHdG were determined by high performance liguid chromatography(HPLC) with fluorescence detection and high performance liquid chromatography-tandem mass spectrometry(LC/MS/MS), respectively. We found that urinary excretion of 1-OHP in coke-oven workers(n=236) is signicantly higher than that in non-exposed workers(n=50). (mean±SD=44.2±61.7μmole/mole cr v.s. 0.62±0.89μmole/mole cr) Our results suggest that 1-OHP is a suitable worker to evaluate the exposure of PAHs. Other results will be presented in the text.
