以相聚合法包覆技術製備紫草萃取液之 微膠囊劑型開發研究

摘要


藉由明膠/阿拉伯膠微膠囊的劑型設計安定當歸和紫草的麻油萃取物。界面活性劑的濃度、明膠濃度和連續相pH值等不同參數對微膠囊粒度分布的影響也包含在本研究中。當界面活性劑/油的比例為1/10,明膠/油的比例為1/5,在pH值4-6時有最佳的相聚合條件。 本研究主要以塑化劑甘油替換最常被用於明膠/阿拉伯膠形成的微膠囊的交聯劑甲醛。粒度分佈分析儀、顯微鏡、掃描式電子顯微鏡、高壓液相層析儀、示差熱分析儀都被使用來研究含紫草素萃取物的明膠/阿拉伯膠形成的微膠囊的物理性質。由顯微鏡的觀察顯示,3%和6%的甘油與0.5%甲醛所形成的單核微膠囊並沒有不同。研究顯示,具有均勻分布及形狀圓形可藉由調整處方得到高達99.6%包埋率的微膠囊,且最多可加入5%萃取物於此處方中,說明甘油可以扮演無毒和省能的塑化劑應用在包埋含紫草素萃取物的微膠囊製備上。 在體外試驗,紫草素萃取物具有輕度的抗胃幽門桿菌作用。還需未來研究來說明此萃取物的藥理作用。

Abstract Extracts of Radix Angelica sinensis and Radix Arnebiae seu lithospermi mixture in sesame oil (shikonin containing extract) is stabilized by the dosage form design in gelatin/acacia microcapsule. The effect of various parameters such as the concentration of surfactant, concentration of gelatin and continuous phase pH condition on the microcapsule particle size distribution was experimentally investigated. It was shown that the optimum concentration for surfactant/oil ratio is 1/10 and gelatin/oil ratio is 1/5 in the pH condition of approximately 4 to 6 for the coacervation process. Modification of the most frequently used crosslinking agent formaldehyde in the gelatin-acacia microencapsulation process was altered by plasticizer glycerol in this study. Physical properties of these microcapsules were obtained from analytical instruments as particle size distribution analyzer, microscopy, scanning electronic microscopy, high performance liquid chromatography and differential scanning calorimetry. Results obtained from microscopy observation revealed that one core microcapsule prepared by 3% and 6% glycerol were no different from 0.5% formaldehyde. The microcapsules had a maximal core material loading content of 5% extract, and high throughput modified process can be reached the encapsulation efficiency of 99.6% with a good dispersion, even distribution, and round shape. Hence, glycerol was demonstrated to be not only a good non-toxic plasticizing material, but also energy saving during microencapsulation process for encapsulated shikonin containing extract applications. In vitro study, shikonin containing extract had moderate in-vitro anti-Helicobacter pylori effect. However, further studies are needed to clarify pharmacological effects for this herbal extract.
