摘 要 台灣青木香(Saussurea deltoidea (DC.) C. B. Clarke)為菊科(Compositae)青木香屬(Saussurea)之多年生草本植物,分布於中國、台灣及喜馬拉雅山一帶中高海拔之山區道路或林道旁。回顧過去研究,僅有大陸地區研究分離得到8 個已知化合物,且尚未有任何活性被報導。因台灣地區台灣青木香之全株甲醇抽出物具有抗發炎活性,因而對全株成分分離及活性研究作探討。 針對台灣青木香全株之甲醇萃取物進行分配萃取後,得到乙酸乙酯層及水層兩個部分,其中乙酸乙酯層顯示具有強抗發炎活性,遂進行乙酸乙酯層的化學成分研究,共獲得24 個化合物,其中有1 個新化合物,23 個已知化合物︰ 一、sesquiterpene 化合物1 個︰deltoidealactone (1),為新化合物。 二、triterpenoids 化合物6 個︰lupeol (2)、betulonic acid (3)、taraxasterol (4)、taraxasteryl acetate (5)、oleanolic acid (6)、β-amyrin (7)。 三、steroids 化合物4 個︰β-sitosterol (8)與stigmasterol (9)之混合物;β-sitostenone (10)與stigmasta-4,22-dien-3-one (11)之混合物。 四、benzenoids 化合物4 個︰vanillin (12)、p-hydroxybenzoic acid (13)、vanillic acid (14)、methyl 4-hydroxybenzoate (15)。 五、quinone 化合物1 個︰α-tocopheryl quinone (16)。 六、alkaloid 化合物1 個:methyl indole-3-carboxylate (17) 七、fatty acids 化合物7 個︰balansenate II (18);stearic acid (19);tetradecanoic acid (20)、pentadecanoic acid (21)及palmitic acid (22)之混合物;methyl palmitate (23)與methyl stearate (24)之混合物。 所有化合物均經資料比對、圖譜解析決定。 deltoidealactone (1)對體外U937 細胞產生TNF-alpha有抑制效果,其抗發炎活性之IC50 值為1.47 ug/mL。
Abstract Saussurea deltoidea (DC.) C. B. Clarke (Compositae) is a perennial herb, and distributed in China, Himalaya and Taiwan. Eight known compounds have been isolated from same species in Mainland China previously. However, the bioactivity of this species has not been conducted. The MeOH extract of Formosan species showed strong anti-inflammatory activities, and the aim of this study is the isolation of chemical constituents and bioactivities of this plant. Investigations of active EtOAc layer of this plant by SiO2 column chromatography led to the isolation of one new compound along with twenty-three known compounds:. 1. one sesquiterpene: deltoidealactone (1), a new compound. 2. six triterpenoids: lupeol (2), betulonic acid (3), taraxasterol (4), taraxasterylacetate (5), oleanoic acid (6), β-amyrin (7). 3. four steroids: a mixture of β-sitosterol (8) and stigmasterol (9); a mixture of β-sitostenone (10) and stigmasta-4,22-dien-3-one (11). 4. four benzenoids: vanillin (12), p-hydroxybenzoic acid (13), vanillic acid (14),methyl 4-hydroxybenzoate (15). 5. one quinone: α-tocopheryl quinone (16). 6. one alkaloid: methyl indole-3-carboxylate (17). 7. seven fatty acids: balansenate II (18); stearic acid (19); a mixture of tetradecanoic acid (20), pentadecanoic acid (21), and palmitic acid (22); a mixture of methyl palmitate (23) and methyl stearate (24). The structures of these compounds were elucidated by spectral analysis. Deltoidealactone (1) inhibits TNF-alpha production in U937 cells in vitro, and the IC50 value is 1.47 ug/mL.
