種子含油量非破壞性磁共振測量暨含油量與麻瘋樹種子性狀相關性之研究

Non-destructive MR method for the measurement of oil content and study of correlation between oil content and growing characteristics of Jatropha seeds.

摘要


本研究主要是利用磁共振分析儀測量法量測有殼、無殼麻瘋樹種子含油量,此低誤差、測量快速又操作簡單的含油量量測方法取代了傳統的索式萃取法,提供了一個兼具可測量單顆種子含油量及非破壞性的測量方法。另一方面,本研究藉由測量700顆種子的重量、長度、寬度和厚度等外觀性狀參數,對含油量作圖並且迴歸,探討長度、寬度、厚度、重量、及密度等參數與種子含油量間的相關性。 最後利用經由磁共振分析儀測量而篩選出的較高含油量種子進行麻瘋樹體外培養,並觀察供給不同氮源濃度時,對麻瘋樹幼苗生長的影響。 結果顯示使用磁共振分析儀測量麻瘋樹種子含油量的誤差僅有0.167%,證實了使用磁共振分析儀測量含油量確實是可行的。而含油量與種子外觀性狀相關性探討的部份,由700個數據與含油量交叉比對後發現,由於種子才是貯油的主要部份,因此相較於含殼麻瘋樹種子含油量還需要額外考慮種殼的長度、寬度、厚度等,無殼麻瘋樹種子的性狀數據皆與含油量呈現較大的正相關。各性狀當中,又以無殼麻瘋樹種子厚度與含油量的相關度最高,相關係數r值為0.3092。而密度參數同時包含了重量參數與體積參數,因此種子的密度與含油量有更高的相關性,有殼麻瘋樹種子密度與含油量的相關係數高達了0.62,顯示了藉由有殼麻瘋樹種子的密度預估種子含油量,進行初步的種子篩選是可行的。實驗最後一個部份由篩選過的高含油量種子進行體外培養,使用不同氮源濃度(0%,50%,100%)培養基進行實驗,結果發現氮源的不足明顯的影響了麻瘋樹幼苗的生長,並且影響了麻瘋樹幼苗葉片中葉綠素的合成,導致氮缺乏的麻瘋樹樹苗葉片為白色或淡綠色且葉片略微乾枯、捲曲,而使用正常氮源劑量培養的麻瘋樹樹苗葉子卻十分健康、翠綠。

In this study, MR is used to exam the oil content of Jatropha curcas seeds. In recent years, traditional Soxhlet extraction method is replaced by MR because of its accuracy, easy operating and simple pretreatment. The most important of all is that it’s a method to exam the oil content without any destruction. Seeds are available to reuse in other application, such as tissue culture. Furthermore, 700 Jatropha curcas seeds with shell and without shell are examined in length, width, thickness and weight for some discussions on the correlation between these characteristics and oil content respectively. The seeds which are selected by NMR as the high content of oil are used in tissue culture to see the effects by giving different nitrogen concentration resource. Data shows that the error value of MR is only 0.167%, which means MR is a reliable way to exam the oil content. After analyzing 700 data, it’s found that the correlation between oil content and seeds characteristics are more apparent for the seeds without shell compared to the seeds with shell. Due to the shells are too heavy and big, the data are barely related to the oil content. However, the correlation between seeds width and oil content is related high, the regression coefficient R value is 0.3092. The correlation between seeds density and oil content is highest among above, because density is a parameter of weight and volume, the R value is up to 0.62, which means we can simply preliminary select seeds oil content by the density of seeds without any examination and extraction. The last part of this study, we can observe that the seedling without any nitrogen resource can’t synthesize the chlorophyll normally, the color of the leaves are white and light green while it’s emerald for those are provided better nitrogen resource.
