摘要


論文摘要 尿酸(Uric acid)是現代高油高脂飲食下易發生的文明病,尿酸是體內嘌呤在肝臟代謝後所形成,會分布於人體的血液與尿液中,而高尿酸血症的患者則易罹患痛風、慢性腎臟病、心血管疾病等。本研究目的為研發一種使用少量待測物即可在短時間內測得人體尿酸含量之陣列型尿酸生物感測器,與其他需大量待測物的傳統檢測方法所造成的不便和花費時間過長比起來,更能達便利、高準確度和及時量測等優點。 本研究是使用三電極的裸金電極當尿酸生物感測器,並在工作電極上固定尿酸酶,提高感測器的靈敏度;藉由循環伏安法量測出感測器氧化電壓為0.9伏,在此電壓範圍下感測器展現極佳的線性度,1Х1單一型生物感測器和1Х2、1Х3陣列型生物感測器之線性度各為0.9798、0.9963和 0.9638。並比較在1.2mM的尿酸標準溶液下的訊噪比和相對標準差,最後,將尿酸生物感測器保存於4℃下,發現其在35天後仍能保存89.23%的原有性能。

Abstract Hyperuricemia is a civilized disease in modern high fat and high oil diet. Uric acid is a product of purin metabolism in the liver, and it is widely distributed in human blood and urine. Patients with hyperuricemia could lead to gout, chronic kidney disease, cardiovascular disease, and etc. An advanced biological sensor is proposed in this thesis. This uric acid array biosensor can immediately detect uric acid for human blood by a little analyte. It is more convenience、accuracy and time-saving in comparison to other traditional detection methods. In this study, the uric acid array biosensor used bare Au electrodes as three electrode system. Next, uricase fixed on the working electrode to promote the sensitivity of biosensor. The uric acid array biosensor showed a good linear response range for determination of uric acid at 0.9V. The linear for determination of uric acid standard solution for 1Х1 uric acid single biosensor, 1Х2 and 1Х3 uric acid array biosensors were 0.9798、0.9963 and 0.9638. The signal-to-noise ratio and relative standard deviations for determination of 1.2mM uric acid standard solution are individually compared. Finally, the uric acid biosensor was stored at 4℃,and still maintained 89.23%from the original activity after 35 days.
