矩陣型肌酸酐生物感測器之製作

摘要


在台灣,根據行政院衛生署統計結果可知,截至2011年腎臟疾病已長達十多年蟬連為國人十大死因之一。因此,慢性腎臟疾病的盛行,已逐漸嚴重危害到國人的健康。肌酸酐已被證實為用來評估腎臟疾病的重要指標之一。本研究的目的為開發一種簡易、創新及高靈敏的矩陣型肌酸酐生物感測器來精準地檢測肌酸酐,而本研究的導納式矩陣型肌酸酐生物感測器,具備了低成本之優點,其利用具有導電性銀膠和碳膠,透過網版印刷技術印刷之線路與電極,再將CD(Creatinine Deiminase)酵素固定於電極上。 我們研究了矩陣型肌酸酐生物感測器之線性度與靈敏度之反應,矩陣型肌酸酐生物感測器在偵測肌酸酐時,表現出極佳的線性反應範圍(50∼250μM)。此外,1x3矩陣型肌酸酐生物感測器的訊號雜訊比(SNR)數值相較於1x1單一型肌酸酐生物感測器高於23%。對於訊號雜訊比的增加,1x3矩陣型肌酸酐生物感測器的靈敏度也有明顯地提升(1x3矩陣型肌酸酐生物感測器靈敏度比起1x1單一型肌酸酐生物感測器高於3倍)。 總而言之,我們研發了一種簡單、低成本與高靈敏度的矩陣型肌酸酐生物感測器,不論是在正常生理及病態的範圍中,都能夠精準檢測血漿中之肌酸酐濃度。

According to the statistics reported by the Department of Health, kidney disease is the people one of the ten leading causes of death in Taiwan. Creatinine is a risk factor for kidney disease. This study aimed to develop a new, simple, low-cost and high sensitivity creatinine biosensor array (CBA) for precise determination of creatinine. The CBA on the detection of creatinine at admittance showed an excellent linear response range (50–250μM). The effects of creatinine biosensor (1x1, 1x2, and 1x3) of the CBA, SNR and sensitivity on the response of the CBA were investigated. The detection sensitivity of the CBA with 1x3 creatinine biosensor could be significantly increased (about 3-fold) as compared to that of the CBA with 1x1 creatinine biosensor because of the increase of signal to noise ratio. Finally, a simple, low-cost, high sensitivity and tiny CBA was developed being able to precisely determine plasma creatinine concentrations in both the normal physiological and pathological ranges.
