摘要


連翹(Forsythia suspensa)為木犀科連翹屬植物。因文獻報導其具有木脂體類化合物,與木脂體類化合物在體內代謝形成mammalian lignans有著相當的關連性,故被選定為研究標的。因此本研究將著重於連翹之木脂體成分研究,希望能發現之更佳前驅物。由連翹甲醇抽取物分離得到22個化合物,其中含量較多的化合物將進一步篩選其抗氧化活性,以評估其是否可以成為新的醫藥資源。 中草藥超微細化能使得藥物顆粒更細小化,對中草藥而言,可節省大量原料成本,大幅提高中藥材的利用率,另一方面亦可增加中草藥劑型的多樣性。超微細化技術將甘草及丹參粉碎至顆粒的大小接近奈米,可以使得主要成分如甘草酸與丹參酚酸B被大量的溶出。腸道細胞(caco-2)進行的穿透測試結果,發現超微細化甘草經由腸道細胞進行穿透測試,穿透液中甘草酸含量結果為0.48 mg/g;而傳統粉碎的甘草穿透液中,甘草酸的含量則低於儀器偵測極限。 由紅色系蝴蝶蘭(Dtps. Tinny Ribbon × Dtps. Plum Rose)的甲醇抽取物共分離鑑定出12個化合物,其中包含2個類黃酮醣苷,1個花青素,及另外9個其他類化合物。在這些化合物中,有1個類黃酮碳苷為首次分離得到的新化合物。利用高效能液相層析法,成功發展出對於多個台灣原生種與商業品種主要花色成分進行定量分析的方法,希望藉此可以加速培育新花色的目的。

Forsythia suspensa (Oleaceae) was reported to contain lignans in the previous reports so that it was selected as the target. Totally twenty two compounds were characterized from the methanol extracts of the F. suspensa and the isolates with more quantities would be subjected into antioxidant activity examination to discover the new medicine resources. Traditional Chinese medicine prepared by ultrafining could make the particles smaller, and it would save the cost and improve the utility rate of the medicine plant materials. In addition, it would provide more choices of the forms of a drug. The particles of Glycyrrhiza uralensis and Salvia miltiorrhiza were grounded to submicroscale by ultrafining, and it resulted in the more release of the major constituents, such as glycyrrhizic acid and salvianolic acid B. The Caco-2 cell lines assay displayed that quantification of the liquid in the basolateral side showed that the amount of glycyrrhizic acid was 0.48 mg/g in the samples prepared by ultrafining, but it was not detectable in the samples grounded traditionally. Two flavone glycosides, one anthocyanidins, and nine other compounds were characterized from the methanolic extracts of Phalaenopsis hybrids (Dtps. Tinny Ribbon × Dtps. Plum Rose). Among these metabolites, one flavone C-glycoside was reported for the first time. Moreover, it was successful to develop quantitative analytical method by HPLC for the major pigments in the Taiwan native and commercial hybrids species. It would be helpful to facilitate the incubation of new colors of Phalaenopsis species.
