陳皮化學成分及生物活性之研究

摘要


陳皮為芸香科 (Rutaceae) 常綠小喬木植物橘 (Citrus reticulata) 的成熟果皮,經乾燥陳放後製成。相關研究顯示柑橘果皮中富含的多甲氧基類黃酮 (polymethoxyflavones; PMFs) 具抗氧化、抗發炎、降低膽固醇等活性。在本研究中,將陳皮以甲醇浸泡萃取,得陳皮甲醇粗萃物,經分配萃取得到乙酸乙酯層、正丁醇層及水層萃取物。在抗氧化結果中,顯示乙酸乙酯層萃取物具有活性。將乙酸乙酯層經由管柱層析分離出三十四組分液,進一步以管柱層析、高效能液相層析與再結晶等方法,進行乙酸乙酯層萃取物純化,經由核磁共振光譜分析與文獻比對,共鑑定出十個化合物,包含三個芳香類 (Aromatics): Vanillin acid、4-Hydroxybenzaldehyde、Ferulic acid;一個檸檬苦素類 (Limonoid): Limonin; 一個固醇類 (Steroid): -Sitosterol;一個香豆素類 (Coumarin):Scoparone;三個黃酮類 (Flavonoids): 3-Hydroxy-5,6,7,8,3′,4′-hexamethoxyflavone、Retusin、3,3',4',5,6,7,8-Heptamethoxyflavone;一個呋喃類 (Furan):5-HMF。 關鍵詞:橘、芸香科、管柱層析、抗氧化、多甲氧基類黃酮

Chen pi is oven- or sun-dried peel of Citrus reticulata used as a traditional Chinese medicine. Many studies demonstrated that polymethoxyflavones (PMFs) from citrus peel possess anti-oxidant, anti-inflammatory and lowering cholesterol effects. In this study, oven-dried peels of Citrus reticulata were extracted with methanol (MeOH). The MeOH crude extract was suspended in water and partitioned with ethyl acetate (EA) and n-butanol (n-BuOH) to obtain the EA, n-BuOH and water layers of Citrus reticulata peels. In the preliminary antioxidant assays, the EA layer of Citrus reticulata peels showed anti-oxidant activity. The EA layer was separated into thirty-four fractions by column chromatography. The fractions were then purified by repeated column chromatography, high performance liquid chromatography (HPLC) and recrystallization to obtain ten compounds, including three aromatics: vanillic acid, 4-hydroxybenzaldehyde, ferulic acid; one limonoid: Limonin; one steroid: -sitosterol; three flavonoids: 3-hydroxy-5,6,7,8,3′,4′-hexamethoxyflavone, Retusin, 3,3',4',5,6,7,8-heptamethoxyflavone; and one furan: 5-HMF.
