月桃(Alpinia speciosa)為薑科(Zingiberaceae)月桃屬(Alpinia)植物,也是台灣野外常見多年生草本植物,其藥理學研究表明具有抗潰瘍、止瀉、利膽、抗炎鎮痛、抗凝血、抗菌作用,另對微循環系统也有影響,依文獻上所敘述之療效及民間長久以來的使用,月桃夾殼應相當具有開發醫療藥物的潛力。 本實驗以2 kg砂仁夾殼甲醇萃取(5 × 5 L)純化後,得到13.14%的粗萃物,經極性分配出氯彷層、正丁醇層、水層。在氯彷層已鑑定出含有0.256 %的二氫-5,6-去氫卡瓦胡椒素(dihydro-5,6-dehydrokawain) (DDK)及0.092% 5,6-去氫卡瓦胡椒素(5,6-dehydrrokawain)。後建立DDK及DK的HPLC分析方法,其檢測濃度範圍分別為40–100和50–140 µg/mL。 測試MeOH、EtOH、10% EtOH-water、water及hexane萃取效果,發現以MeOH、EtOH、Hexane萃取夾殼DDK及DK較好,其含量分別為0.52 %、0.48 %、0.54 %及0.20、0.20、0.21%,其中以Hexane萃取效率最好,可以得到高純度DDK及DK。再以索氏萃取比較砂仁及夾殼DDK及DK的含量,數據表明以夾殼有較好之DDK (0.54%)及DK (0.21%)產率。因此,以夾殼為材料大量製備DDk及DK等成分並且簡化萃取流程,期望能增加月桃夾殼的經濟價值。
Alpinia speciosa (shell Ginger, Syn: A. zerumbet), (Zingiberaceae) is a perennial herb traditionally used in Taiwan for the treatment of ulcers, diarrhea, gallbladder, inflammatory, and analgesic complications. The purpose of this study is to identify the potential bioactive compounds from fruit shells of A. speciosa. The fruit shells (2 kg) were extracted with methanol (5×5L) under reflux. The combined methanol extract (267.0 g, 13.4%) was concentrated and dissolved in water and, then portioned with chloroform (127.9 g, 6.4%), n-butanol (21.9 g, 1.1%) and water (118.1 g, 5.9) fractions successively. The chloroform extract was purified by silica gel column chromatography using hexane/ethyl acetate gradient to afford dihydro-5,6-dehydrokawain (DDK, 5.12g, 0.256%) and 5,6-dehydrrokawain (DK, 1.84 g, 0.092%). The HPLC standard curves were established for DDK and DK in the concentration range 40–100 and 50–140 µg/mL, respectively. On the other hand, fruit shells of A. speciosa were extracted under reflux with MeOH, EtOH, 10% EtOH in water, water, and hexane to know the best solvent to obtain maximal DDK and DK with high purity. The HPLC analysis revealed that though MeOH, EtOH, and hexane extracts gave nearly similar DDK (0.52, 0.48 and 0.54%, respectively) and DK (0.20, 0.20 and 0.21%, respectively) yields, however hexane extract resulted the high purity of DDK and DK. These results suggest that hexane is a favorable solvent to get the high purity of DDK and DK from fruit shells of A. speciosa. Furthermore, hexane extract of fruit shells resulted the high yield of DDK (0.54%) and DK (0.21%) compared with seeds with a yield of 0.012 and 0.007%, respectively. The neuroprotective activities of DDK and DK are interesting and, studies are under progress. Thus our study may be useful to get the high yield bioactive DDK and DK from A. speciosa.
