石蓮花為一種多年生草本植物,屬於景天科 (Crassulaceae)石蓮花屬 (Graptopetalum) 。其中發現景天科類植物具有以下功能:活化細胞、防止肝硬化、降血壓、血糖及高尿酸、血栓發生率、治療痛風、 軟化血管、抗發炎、抗菌及抗病毒等功能。因此本研究希望從石蓮花組織的萃取液中找尋抗凝血物質。由石蓮花初萃取之PT (Prothrombin time)及APTT (Activated partial thromboplastin time)結果顯示石蓮花的萃取液確實具有延遲血液凝固的功能,平均為正常凝血時間的數倍,且分佈於葉皮 (PT:2.1倍,APTT:1.3倍)、葉肉 (PT:2.6倍,APTT:2.3倍)及全葉 (PT:2.6倍,APTT:2.2倍)等組織。石蓮花抗凝血物質純化上,將石蓮花乾燥回溶液經過高壓濃縮、G-25管柱膠體層析、HPLC Hypersil GOLD管柱等純化步驟後,純化倍率可達4.7倍,殘存活性約有40%。純化物質特性分析上,G-25管柱膠體層析分析,純化物質分子量小於500 Da。元素分析,顯示碳含量為24.19%、氫含量為3.055%、硫含量為0.258%。FT-IR分析,顯示含有羥基 (-OH)、苯環 (Aromatic C=C)、酮基 (C=O)等官能基。醣類及蛋白質鑑定分析,顯示此純化物質可能不是醣類或蛋白質。熱穩定度上,PT試驗中,至100℃仍有活性。而APTT試驗中,至60℃以上即喪失活性。
Graptopetalum paraguayense is a perennial herbaceous plant that belongs to Crassulaceae subject and Graptopetalum genus. some Crassulaceae subject could be used for the following functions: cell activation, liver cirrhosis prevention, blood pressure reducing, blood sugar reducing, thrombus formation rate reducing, gout counteract therapy, vessel softening, anti-hyperuricemia, anti-inflammation, antibiotic, and also anti-virus. In this study, we hoped to find the anticoagulant which obtained from G. paraguayense leaf tissue extract. By the result of extract from G. paraguayense for PT test (Prothrombin time) and APTT test (Activated partial thromboplastin time) showed that its extract really postponed the blood coagulation about several folds of the normal blood coagulation on average. And it was distributed over the leaf epidermis (PT : 2.1 folds ; APTT : 1.3 folds), mesophyll (PT : 2.6 folds ; APTT : 2.3 folds), full leaf (PT : 2.6 folds ; APTT : 2.2 folds). In the purification of G. paraguayense anticoagulant, dried extract was dissolved into double distill water (ddH2O). Followed by high-pressure concentration, G-25 chromatography, and HPLC Hypersil GOLD column; a substrate of 4.7 purification folds as well as the survival activity of 40% was obtained. G-25 chromatography analysis revealed that the molecular weight of this anticoagulant substrate was lower than 500 Dalton. The elemental analysis revealed that it had 24.19% carbon, 3.055% hydrogen, and 0.258% sulfur. The FT-IR (Fourier transform infrared spectrometer) showed the substrate contains hydroxyl, ketone, and aromatic functional group. After glycoside and amino acid analysis were processed, this substrate of interest is neither sugar nor protein. The temperature stability of this substrate, it retains activity up to 100℃ in PT test but loses activity when heated more than 60℃ in APTT test.
