嗜鹽菌Haloferax mediterranei為紅色色素的生產菌株,其細胞膜含有菌紅素(Bacterioruberin),是一類不常見的高碳數羥基化C50類胡蘿蔔素,具有抗氧化性等應用價值。本研究的目的乃是在利用液-液相萃取法,探討培養在極高鹽度環境下的H. mediterranei菌體內紅色素萃取物(Extracted Red Pigment from H. mediterranei, hmERP),並運用高效能液相層析儀(High Performance Liquid Chromatography, HPLC)結合電噴灑串聯質譜儀(Electrospray ionization Tandem Mass Spectrometer, ESI-MS-MS)與霍氏轉換紅外光譜儀(Fourier Transform Infrared spectrophotometer, FTIR)進行成份與化學結構之研究。hmERP經FTIR吸收光譜顯示可能官能基於3468㎝-1為O-H伸縮振動;於2964㎝-1與2963㎝-1為亞甲基(-CH2-)的吸收;於1673㎝-1為共軛雙烯的伸縮振動(C=C)。而ESI-MS-MS於數據處理方式採用選擇性離子檢測(Selective ion monitoring)模式下所得分子離子峰的數據與裂解訊息,推算hmERP內含分子之可能分子式,並建立簡便的指紋特徵圖譜以提供hmERP色素的鑒別。
Haloferax mediterranei produces a mixture of red pigment in which Bacterioruberin, an unusual membrane pigment belong to hydroxyl C50 carotenoid that shows an anti-oxidant property. The purpose of this study is to investigate the extracted red pigments from H. mediterranei (hmERP) under environments of extremely high concentration of salts. Then to study the composition and also the chemical structure of hmERP by high performance liquid chromatography (HPLC) Combining with the electrospray ionization tandem mass spectrometer (ESI-MS-MS), and also by Fourier transform infrared spectrophotometer (FTIR). The FTIR spectrums show that several chemical structure in the mixture of hmERP, that are O-H flexible vibration at 3468 cm-1, inferior methyl (-CH2-) absorption at 2964 cm-1 and 2963 cm-1, and vibration of a pair of alkene of conjugation (C=C) at 1673 cm-1. The chemical structures of different constituents in hmERP were determined by ESI-MS-MS based on the fragmentation assignment and molecular weights, and also established the characteristic fingerprint of major components in hmERP with selective ion monitoring (SIM). This method is simple, rapid and can be used for identification of characteristic fingerprint.
