紅柿葉萃取物的抗氧化功能分析與應用

摘要


大部份染髮劑主要的成份為對苯二胺類化合物(p-Phenylenediamine)、過氧化氫(Hydrogen peroxide)、酚類化合物(Phenolic compounds)等氧化劑。上述這些化學藥劑,對人體的泌尿系統、淋巴系統、神經系統造成不同程度的皮膚紅斑或水腫等過敏現象。染髮劑的化學成份從毛囊滲透到血液,並在人體內積存。長期使用染髮劑會使致癌風險增加。本研究使用中草藥萃取物為紅柿葉,其成分含有多酚類(Polyphenol)、類黃酮(Flavonoid)及維生素(Vitamin)等。希望藉由紅柿葉萃取物的抗氧化功能,降低染髮劑對頭皮的傷害。以甲醇泡製將紅柿葉過濾後,經冷凝減壓萃取及冷凍乾燥得到萃取物。將紅柿葉萃取物進行2,2-二苯基-1-苦基肼基 (2,2-diphenyl-1-picrylhydrazyl, DPPH) 清除自由基能力、人類皮膚纖維母細胞(Human skin fibroblast cell, CCD-966SK)毒性試驗、降低過氧化氫與對苯二胺對細胞傷害試驗。研究結果顯示0.78 mg/ml紅柿葉萃取物具有抗氧化能力、最小細胞毒性及具有保護細胞並降低過氧化氫及對苯二胺的傷害。

The major ingredients in hair dyes includes p-phenylenediamine compounds, hydrogen peroxide and phenolic compounds. The about chemical compounds may affect the urinary system, lymphatic system, nervous system, and cause skin erythema or allergic edema. These chemical compounds penetrate into the blood from the hair follicle, might increase the risk of cancer. The Chinese herbal extract used in this study is red persimmon leaf. The extract contains polyphenols, flavonoid and vitamins, etc. The red persimmon leaves were soaked with methanol, and then extracted by condensation and vacuum freeze-drying extraction. The red persimmon leaf extract was characterized by DPPH scavenging test, cytotoxicity assay, and hydrogen peroxide and p-phenylenediamine damage test. The results showed that persimmon leaf extract at the concentration of 0.78 mg/ml obtained high antioxidant ability, no cytotoxicity, and also protect cells from hydrogen peroxide and p-phenylenediamine injury. We hope to protect the scalp from chemical damage by utilizing the antioxidant function of persimmon leaves extract.
