綠竹葉萃取液之抗氧化活性分析及其美容應用之評估

A Study of the anti-oxidative Activities of Green Bamboo Leaves Extract and Its Application in Cosmetic Formulations

摘要


本研究主要探討綠竹葉萃取液的抗氧化活性,及其美容應用之評估。在抗氧化活性試驗中,以新鮮綠竹葉為材料,探討其10%乙醇溶液萃取物經冷凍乾燥後之抗氧化活性。抗氧化活性檢測項目包括DPPH自由基清除能力、總還原力、亞鐵離子螯合能力、H2O2清除能力、超氧陰離子自由基清除能力及類黃酮含量(Flavonoid contents)等。實驗結果顯示,綠竹葉的粗萃取液具有顯著的抗氧化活性,以DPPH自由基檢測綠竹葉粗萃取液的自由基清除能力,其IC0.20為9.68±0.02Ug/m L。總還原力測試,綠竹葉萃取液的作用力約為維生素E之30%。螯合亞鐵能力之測定結果顯示綠竹葉之螯合金屬能力為檸檬酸之25%。H2O2清除能力測定結果顯示,綠竹葉萃取液無法清除H2O2。此外,綠竹葉也可以清除由xanthine/xanthine oxidase所產生之超氧陰離子自由基(superoxide anion radical)。而其粗抽物之類黃酮含量為50%。因此,由實驗結果顯示,綠竹葉萃取液中含有清除自由基的天然活性成分。同時,本實驗還利用人類的正常皮膚細胞進行試驗,以MTT assay做細胞生長活性檢測。結果顯示,綠竹葉萃取液對皮膚角質細胞及纖維母細胞有促進增生之作用。綜合以上結果,綠竹葉萃取液具有顯著的抗氧化活性,將來可能用以開發為新的抗老化化粧品活性成分。

The aim of this study is to investigate the anti-oxidative activities of green bamboo leaves extract and its application in cosmetic formulations. The anti-oxidative properties of 10% ethanol extracts of bamboos including Bambusa lodhamii Munro (BLM) have been evaluated by DPPH assay, reducing power assay, ferrous ion chelating ability assay, H2O2 clearance assay, superoxide anion scavenger assay, and flavonoid contents assay. The 10% ethanol extracts of bamboos leave showed good performance in all the assays except H2O2 clearance assay. Both the DPPH radical (IC0.2: 9.68+0.02) and superoxide anion radical generated by xanthine/xanthine oxidase system could both be scavenged by the 10% ethanol extracts of green bamboo leaves. The results indicated that Bambusa lodhamii Munro (BLM) extract has 30% of the reducing power of vitamin E and 25% of that of citric acid evaluated by the ferrous ion chelating ability test. The flavonoids contents test result was 50 μg/l00 μg-crude extract. It is concluded that Bambusa lodhamii Munro (BLM) extract contains natural anti-oxidative ingredients. The cell viability assay by MTT methods also has been conducted. The primary data showed that Bambusa lodhamii Munro extract could promote the skin cell proliferation of epidermal keratinocyte and dermal fibroblast without cell cytotoxicity. Based on the anti-oxidative property of Bambusa lodhamii Munro extracts, we speculated that it may have anti-aging potential. The future work is to identify the antioxidant components in Bambusa lodhamii Munro, which can also promote skin cell proliferation. We believe that the experiment result would contribute to the anti-aging cosmetic technology.
