Preliminary Periodontal Macrophage Sorting for HCMV Infection Investigation

人類巨細胞病毒與牙周感染之關係之先遣研究-牙周巨噬細胞之分離

摘要


在現今許多與嚴重型牙周病(severe periodontal disease)相關的研究中,由於人類巨細胞病毒(human cytomegalovirus, HCMV)其生命週期(life cycle)和牙周疾病發病的週期相當類似,因此對嚴重型牙周病而言,人類巨細胞病毒可能是一項相當重要的致病因子。根據相關的醫學文獻,在眾多細胞中,單核球細胞(monocyte)與巨噬細胞(macrophage)是人類巨細胞病毒最主要感染的目標,因此牙周巨噬細胞(periodontal macrophage)在受人類巨細胞病毒感染後其所表現的促發炎細胞激素(pro-inflammatory cytokine)與牙周破壞之間的關係是目前我們相當感興趣的一個研究主題,但在國科會大專生研究計畫有限的經費之下,我們初步先研究自發炎的牙齦組織中分離出牙周巨噬細胞的方法,以期將來能更進一步用於研究人類巨細胞病毒與嚴重型牙周病之關係。

Human cytomegalovirus (HCMV), a herpesvirinae, is one of largest, enveloped virus containing doublestrand DNA that infect humans. Many researches revealed that it may play a role in etiopathogenesis of several severe periodontal disease and parts of its life cycle are similar to disease progression. Among all host cells of HCMV, medical literature reports that monocyte/macrophage are the main target cells of HCMV infection. Therefore, the aim at this research is intended to identify the actual role of expression of pro-inflammatory cytokines associated with periodontal breakdown of in Vitro HCMV-infected periodontal macrophage. However, within the limited research fund of NSC for undergraduate students, we initially propose a preliminary project to establish a working chart of culturing CD14 labeled periodontal macrophages specifically for further periodontal associated HCMV research.
