梔子化學成分之HPLC分析

Studies on Chemical Components of Gardeniae Fructus by HPLC

摘要


梔子(Gardeniae Fructus)為常用中藥之一,《神農本草經》列為中品,中華中藥典規範其基原為茜草科(Rubiaceae)植物梔子Gardenia jasminoides Eills的乾燥成熟果實。常誤用藥材為水梔子Gardenia jasminoides Eills var. granidiflora Nakai,雖為梔子之變種植物,但其外觀性狀、組織與成分含量仍有一定差異,應予區別。有鑑於此,本研究為了解市售梔子藥材之使用情形,搜集梔子對照藥材,由外觀性狀特徵及薄層層析法加以確認基原,再利用高效液相層析法,針對其指標成分geniposide進行定量。結果發現市售21件檢體均為梔子正品,其geniposide含量為4.12-7.13%,符合中華中藥典規範geniposide含量不得低於2%之規範,而建立之高效液相層析方法,可不受其他成分干擾,對3種梔子之指標成分有良好的分離效果。

The botanical origin of Gardeniae Fructus is fruit of Gardenia jasminoides Eills. According to literatures, G. jasminoides var. grandiflora is an alternative. Twenty-one samples were purchased from market and were distinguished from authentic material by morphology, and thin layer chromatographic analysis. In addition, the amount of geniposide was determined by high-performance liquid chromatography. The results showed that all samples were G. jasminoides. The amounts of geniposide in those samples ranged from 4.12 to 7.13%. According to Chinese pharmacopoeia Gardeniae Fructus shall contain not less than 2% of geniposide, as the result, all samples were qualified for the specification. Three marker compounds, geniposide, gardenoside and geniposidic acid of Gardeniae Fructus could also be separated by this HPLC method.
