花生幼齡子房柄的體外培養Ⅲ、添加酪素水解物的影響及其與植物生長調節劑之交互效應

In vitro Culture of Peanut (Arachis hypogaea L.) Young Pegs Ⅲ. Influence of Casein Hydrolysate and the Interactions with Plant Growth Regulators

摘要


本研究利用栽培種落花生台南11號之自花授粉後7天的幼齡子房柄為培殖體,探討添加酪素水解物(casein hydrolysate, CH)及植物生長調節劑對英果形成與種子發育的影響。結果摘述如下: 1. MS添加150mg/l CH培養基的莢果形成率為76.7%,與單純MS培養基的莢果形成率(78.3%)無顯著差異,但莢果形成率隨CH添加量增加而皇降低的趨勢。 2.在含CH的培養基中加添NAA雖可降低停止生長及僅誘導癒傷組織的比率,但子房柄多僅呈伸長反應,對莢果形成並無任何助益。添加BA促進癒傷組織生成,但提高未發根結莢率。同時添加NAA及BA對子房柄形成癒傷組織及不發根結莢有加成的促進作用,但對發根結莢則有加成的抑制作用。 3.添加0.5mg/l的GA可降低停止生長、僅誘導癒傷組織及僅呈伸長反應的比率,對莢果形成則有促進作用。子房柄經MS+150mg/l CH+1mg/l NAA+0.05mg/l BA +0.5mg/l GA的培養基培養120天有高達94.3%的莢果形成率。但CH提高為300mg/l則僅有29.2%之莢果形成率。 4.就莢果發育而言,單獨添加CH或植物生長調節質問產生大於4mm胚珠的比例均不高;於MS+150mg/l CH的培養基中,添加1mg/l NAA和0.05mg/l BA或添加1mg/l NAA、0.05mg/l BA和0.5mg/l GA,分別有13.6%及10.0%的胚珠寬度大於4mm,其中有可以正常發芽的成熟種子。

The research used young peg tips which were 7 days after pollination of TN 11 as explants, to investigate the responses of pod formation and seed development by adding case hydrolysate (CH) and plant growth regulators. Results were summarized as follows: 1. The pod formative percentage of MS adding 150 mg/l CH medium was 76.7%, which had no significant difference with the 78.3% pod formative percentage obtained by using pure MS medium; however, adding CR decreased the pod formative percentage. 2. Although adding NAA in MS+CH media could reduce the percentage of retention growth, promoted callus formation and promote more peg tips elongation, there was no improvement in the pod formation. Adding BA in MS+CH media raised callus formation and the percentage of the podding without root. Adding both NAA and BA to medium would promote the percentage of callus formation of tip and podding without root, but would inhibit the podding with root. 3. Adding 0.5 mg/l GA could decrease the percentage of stopped growth, of callus formation and of peg tip elongated, but promote the pod development. When the peg tip was cultivated in MS+150 mg/l CR+1 mg/l NAA+ 0.05 mg/l BA+0.5 mg/l GA medium for 120 days, 94.3% of pod were formed. When CR was raised to 300 mg/l, only 29.2% of peg tips will become the pod. 4. For pod development, media containing CH and plant growth regulator produced the lower proportion of ovules larger than 4 mm. The MS+150 mg/l CH medium adding 1 mg/l NAA and 0.05 mg/ l BA or adding 1 mg/l NAA, 005 mg/l BA and 0.5 mg/l GA, obtained 13.6% and 10.0% ovules larger than 4 mm, individually, and some germinated seeds have been observed.
