十種中藥材之自由基清除與酪胺酸酶抑制能力評估

Effects of Selected Ten Traditional Chinese Herbal Medicines on Radical Production and Tyrosinase Activity

摘要


本實驗參考『神農本草經』,挑選女萎、桃仁、白芷、柏實、辛夷、絡石、敗醬、甘草、積雪草與黃芩等十種與美容相關的中藥材,比較其DPPH自由基清除率、TEAC總抗氧化能力、總多酚與總黃酮含量多寡找出較具潛力之藥材,再對有潛力之藥材建立其HPLC之成分指紋圖譜並進行酪胺酸酶抑制率試驗。研究結果顯示,在抗氧化分析方面,各植物水萃物之凍乾樣品在清除DPPH自由基能力之結果,抗氧化力隨著樣品濃度之增加而增加,黃芩與絡石有最佳的清除率,在濃度爲200μg/mL時均超過50%;TEAC值測定結果發現絡石、黃芩與辛夷的抗氧化力效果在濃度爲50μg/mL時均超過95%;在總多酚定量試驗,絡石與黃芩的每克水萃物相當於191.89mg與125.72mg之Gallic acid。總類黃酮測定,絡石與黃芩之每克水萃物相當於29.34mg與70.23mg之Rutin。以HPL作絡石指紋圖譜成份分析,顯示含有gallic acid、chlorgenic acid、caffeic acid、taxifolin、isoquercitrin與quercetin等成分,酪胺酸酶活性評估顯示黃芩與絡石水萃物在3000 μg/mL時,其抑制率均可達60%以上。綜合言之,黃芩與絡石爲較具抗氧化與美白潛力之中藥材,可應用化妝品材料開發。

This study was aimed to evaluate the antioxidative activities of ten herbal plants used for skin-care from ”The Herbal Classic of the Divine Plowman”, including Polygonatum odoratum, Prunus persica, Angelica dahurica, Platycladus orientalis, Magnolia denudate, Trachelospermum jasminoides, Patrinia villosa, Glycyrrhiza uralensis, Centella asiatica and Scutellaria baicalensis. The antioxidative activities assays include the scavenging effect on α, α-diphenyl-β-picrylhydrazyl (DPPH) radicals, the trolox equivalent antioxidant capacity (TEAC), total polyphenols and flavonoids. The HPLC-UV finger print chromatogram and antityrosinase effects of the potent herbs were also measured. The results showed that all extracts obtained antioxidative abilities and their effects depended strongly on their concentrations. Trachelospermum jasminoides and Scutellaria baicalensis had the greatest DPPH radical scavenging effect (both were above 50% at 200μg/mL) and Trachelo5pennwnjasminoides, Scutellaria baicalensis and Magnolia denudate had good antioxidative activity by TEAC (all were above 95% at 50μg/mL). Trachelospermum jasminoides and Scutellaria baicalensis had total polyphenol contents (191.89 mg and 125.72 mg of gallic acid, respectively) and total flavonoid contents (29.34 mg and 70.23 mg of rutin, respectively). Both exhibited potent inhibitory effects on tyrosinase (both were above 60% at 3000μg/mL). A high performance liquid chromatography analysis revealed that phenolic acids and flavonols such as gallic acid, chlorgenic acid, caffeic acid, taxifolin, isoquercitrin and quercetin were present in Trachelospermum jasminoides. Among these ten herbs, Trachelospermum jasminoides and Scutellaria baicalensis were especially potent in terms of antioxidative activities and whitening effects, making them the strongest candidates for cosmetic application found in the current study.
