利用黃水茄(Solanum incanum)果肉細胞培養生產solamargine

摘要


黃水茄(Solanum incanum)果實大小約1~2 cm之果肉為培植體,以MS為基本鹽類添加NAA及BA的培養基可誘得生長快速的癒合組織。在液態懸浮培養中,細胞培養至第18天時細胞鮮重可達到3.88 g/ml而solamargine量則可達到150.04 mg/g.DW。在液態懸浮培養中加入jasmonic acid(2 mg/l、4 mg/l、6 mg/l),皆可促進solamargine在細胞中的累積量,其中以添加2 mg/l jasmonic acid於液態培養基中可獲得最高的細胞增殖(細胞鮮重增加約11倍)量及solamargine產量(206.09 mg/g.DW)。 利用搖瓶進行放大培養,以階段式的培養可獲得較佳的細胞增殖量及solamargine產量,培養18天後,細胞量由原先13.01 g增加為36.01 g而solamargine於細胞中的含量高達306.21 mg/g.DW。

The fastest growing calli were induced from fruits pulp (1~2 cm ) explant of Solanum incanum on MS basal medium contained with NAA plus BA. In cell suspension culture, cell fresh weight achieved 3.88 g/ml and solamargine accumulation 150.04 mg/g.DW when cell cultured in MS+0.2 mg/l NAA+2.25 mg/l BA after 18 days culture. Addition jasmonic acid (2 mg/l, 4 mg/l, 6 mg/l) to liquid suspension medium significantly promote solamargine accumulation in cell and jasmonic acid at 2 mg/l showed the best cell multiplication (biomass up to 11-fold) and solamargine accumulation 206.09 mg/g.DW. Cell was scaled up by flask. In a two stage culture obtained the best cell multiplication and solamargine accumulation. After 18 days, fresh weight of cell was increased from 13.01 g to 36.01 g and the accumulation of solamargine in cell was 306.21 mg/g.DW .
