為了改良葉綠素萃取方法,本研究嘗試利用超音波法配合丙李玉酮、DMF(Dimethylformamide)及DMSO(Dimethyl sulfoxide)三種有機溶劑萃取樟樹葉片之葉綠素,然後以紫外-可見光光譜儀分析,評估萃取方法與萃取溶劑對萃取效果之影響,並探討光譜曲線之變化,同時計算葉綠素之含量。試驗結果顯示,最佳的萃取方法係將葉片之粉末(小於0.7mm)置於DMF溶劑中,並以超音波法取代傳統研磨法,萃取3分鐘後即可。此法操作簡易且萃取快速,除可獲得較佳的萃取效果外,並可增加試驗分析的準確性。與葉綠素在丙酮溶劑中的光譜特性吸收峰位置相比較,葉綠素的光譜特性吸收峰在DMF與DMSO兩種溶劑中顯現了向紅位移。以DMSO、80%丙酮與DMF三種溶劑萃取出來之葉綠素含量分別為l.765mg/g、2.061mg/g與3.628mg/g。
The purpose of this study is to establish a simple method for chlorophyll extraction using ultrasonics. The leaf chlorophyll of Cinnamomum camphora was extracted with acetone, DMF, or DMSO using ultrasonics. The chlorophyll extracts were analyzed with a UV-Visible spectrophotometer. The spectroscopic characteristics of 3 chlorophyll extracts were compared and chlorophyll content was quantified. The experimental results demonstrate that extraction of leaf powders (particle diameters smaller than 0.7mm) with DMF by ultrasonics for 3 min is a rapid, convenient, and effective method for extracting chlorophyll. Compared with the absorption positions of chlorophyll in acetone, a bathochromic shift was observed at the absorption peaks of chlorophyll in DMF and DMSO. The chlorophyll contents extracted with DMSO, 80% acetone, and DMF were 1.765mg/g, 2.061mg/g, and 3.628mg/g, respectively.
