摘要


以多套式聚合酵素鏈鎖反應(multiplex PCR)方法同時檢測五種重要食品中毒原因菌,包括仙人掌桿菌(Bacillus cereus)、沙門氏桿菌(Salmonella spp.)、大腸桿菌O157:H7 (Escherichia coli O157:H7)、單核球增多性李斯特菌(Listeria monocytogenes)及腸炎弧菌(Vibrio parahaemolyticus)。每一對引子對於特定菌株之目標基因具有專一性,其偵測極限分別為仙人掌桿菌10 pg、沙門氏桿菌10 pg、大腸桿菌O157:H7 1ng、單核球增多性李斯特菌100 pg及腸炎弧菌1 ng,並且對於24株非目標菌株不會有特異性片段檢出,本研究利用multiplex PCR可同時快速準確檢測五種食品中毒原因菌。

A multiplex polymerase chain reaction (PCR) method was designed for the simultaneous detection of the five important pathogens, Bacillus cereus, Salmonella spp., Escherichia coli O157:H7, Listeria monocytogenes, and Vibrio parahaemolyticus. Each of the five pairs of primers exclusively amplified the targeted gene of the specific microorganism. The detection limits of the method were 10 pg, 10 pg, 1 ng, 100 pg, and 1 ng for Bacillus cereus, Salmonella spp., Escherichia coli O157:H7, Listeria monocytogenes, and Vibrio parahaemolyticus, respetively. Multiplex PCR did not produce any nonspecific amplification products when tested against 24 nontargeted species. Therefore, the multiplex PCR assay could be a rapid and reliable diagnostic tool for the simultaneous detection of the five pathogenic bacteria.
