菊花萃取物於抗氧化、抑制黑色素生成及抗致突變的研究

Study of Chrysanthemum morifolium Flowers Extract on the Efficacy of Antioxidation, Inhibiting the Formation of Melanin and Antimutagenicity

摘要


本研究採用菊(Chrysanthemum morifolium Ramat.)的乾燥頭狀花序為材料,經95%乙醇萃取後,所製得的菊花乙醇萃取物再進行抗氧化性、抑制黑色素生成的效果以及抗致突變性等試驗。在抗氧化方面,10.0 mg/mL菊花乙醇萃取物清除DPPH自由基的效果相當於同濃度Ascorbic acid的86.4%、BHA的86.3%以及BHT的91.0%;10.0 mg/mL菊花乙醇萃取物的還原力相當於同濃度Ascorbic acid的95.4%、BHA的93.3%以及BHT的101.0%;1.0mg/mL菊花乙醇萃取物螯合亞鐵離子的效果相當於同濃度EDTA的76.3%;1.0 mg/mL菊花乙醇萃取物清除超氧陰離子的效果相當於同濃度BHT的52.0%。在抑制黑色素生成方面,菊花乙醇萃取物抑制生成黑色素的中間產物Dopachrome的IC50為0.76 mg/mL。另由沙門氏菌恢復突變性試驗之結果顯示菊花乙醇萃取物不具致突變性,而且1.0 mg/plate菊花乙醇萃取物對4-NQO引起致沙門氏菌TA100突變的抑制率為79.19%,屬於高度活性的抑制作用(抑制率≧60%)。綜合上述結果,菊花乙醇萃取物應可考量作為抗老化、抑制黑斑,且為一安全而具有化學防護效果的天然保健素材。

The dried flowers of C. morifolium were extracted by 95% ethanol, the ethanolic extract was examined on the effects in the antioxidative activity, the inhibiting the formation of dopachrome and the antimutagenicity. In the concentration of 10.0 mg/mL, the ability of the ethanolic extract for the scavenging DPPH free radicals is 86.4%, 86.3% and 91.0%, while, for the reducing ferric ion is 95.4%, 93.3% and 101.0% to Ascorbic acid, BHA, and BHT, respectively. In 1.0 mg/mL, the ability for the chelating ferrous ion is 76.3% to EDTA. In addition, in 1.0 mg/mL, the ability for the scavenging superoxide anion radicals is 52.0% to BHT. The results indicate the ethanolic extract behaves antioxidative activity. The IC50 of the ethanolic extract for the formation of dopachrome is 0.76 mg/mL. Indicating that the ethanolic extract shows the activity for inhibiting melanogenesis. The ethanolic extract does not possess mutagenicity. The suppressing rate of the ethanolic extract with 1.0 mg/plate for the mutation of Salmonella typhimurium TA100 caused by 4-NQO is 79.19% which is higher than 60%. Suggesting that the ethanolic extract belongs to the strong activity group of antimutagenicity. In conclusion, the ethanolic extract of C. morifolium flowers may be a safe, natural, and healthy material for preventing aging, whitening skin and keeping genes in cells from chemical injury.
