Chemical Strategies for Peptide Synthesis: Amide Bond Formation and Chemical Ligation

摘要


蛋白質在生物體中扮演不可或缺的角色,如幫助代謝以維持生物體機能,或是扮演抵禦外來病菌的免疫系統功能。因此,了解這些蛋白質在生物體內的構型及其作用機制成為後基因體時代重要的任務。然而,有些蛋白質在生物體內的含量低且不穩定,因此不容易取得以進一步研究。所以科學家們希望能藉由體外合成的方式來合成這些重要的蛋白質來進行研究。在本篇文章中,將會從組成蛋白質最重要的鍵結:醯胺鍵開始講起,接著會介紹如何以高效率的化學鏈結方式合成重要之長鏈胜肽分子。

Proteins play important roles in living organisms, such as involving in metabolism for maintaining the essential function and participating in a variety of biological pathways. Therefore, understanding of its biological function, location and binding associates becomes an very important issue for advanced studies. However, the instability as well as low expression level makes certain important proteins very difficult to be collected. Therefore, peptide synthesis and peptide ligation may promise an avenue for preparing those polypeptide chains with reasonable yields and purity. In this review, we try to introduce the background of peptide synthesis and further extend to recent progress of peptide-peptide ligations.
