摘要


含有0.1~0.5mg/1 Naphthalereacetic acid (NAA),0.1~0.5mg/1 Furfurylaminopurine (Kinetin)及2.5% Sucrose之Murashige et al. (1972)基本鹽類培養基可誘導蘆筍培養莖頂形成最多之芽及根。田間採取不同質地,不同位置之芽體培養顯示白色硬莖芽體之繁殖力優於綠色軟莖,距頂芽1cm以內之芽體又優於遠離頂芽者。不同品種間及品種內不同單株間莖頂培養誘導芽體繁殖之能力差異不大。莖頂培養大量繁殖後之試管苗,於11月份低溫下移植之成活率高於5月份高溫移植者。移植之最適苗齡爲培養後3個月之試管苗,配合低溫及適當保濕,其成活率可達87%。莖頂培養繁殖之植株,經與母株外型比較及根端細胞染色體檢定,均顯示與母株相同,證明莖頂培養是大量繁殖優良母株的最好方法之一。

Asparagus (Asparagus officinalis L.) shoot tips of UC157 and UC309 were cultured on Murashige et al. (1972) basal medium supplemented with various am) unts of auxin and cytokinin. It was found that the basal medium added with 0.3 mg/l NAA, 0.1mg/l kinetin and 2.5% sucrose could provide the best result in producing complete plantlets from the excised shoot tips. The growth and differentiating ability of buds from the white hard spear (cv. UC309) was superior to that from the green soft spear. Furthermore, bud section within 1 cm from the shoot tip showed the highest growth vigor and differentiating rate than the lower bud sections. No difference in the ability of shoot production was found among individual plants of th sane variety or among different varieties. Survival rate of asparagus seedlings transplanted from test tube to the greenhouse was higher when performed under low tem2erature (November) than under high tempe rature (May). Seedling age of three-month in test tube was considered the most suitable for transplanting. When high humidity was maintained, survival rate as high as 87% could be obtained. Cytological examination on the root tips of asparagus seedlings originated from shoot tips or lateral buds of spear culture showed diploid chromosome number of 2N=20. No polyploid plants were found. Those results indicated that this propagating method can effectively maintain the nature of mother plants.
