固定幾丁聚醣聚丙烯不織布在抗菌醫療敷材的應用

Immobilized Chitosan onto Polypropylene Non-Woven Fabric for Anti-bacterial and Biomedical Applications

摘要


本實驗是利用低溫高密度氧氣微波電漿活化聚丙烯不織布,使其表面產生高度的氧化基團,隨之接枝丙烯酸單體,使表面具官能基與親水性的結構;再以1-ethyl-3-(3-dimethy-laminopropyl) carbodiimide hydrochloride (EDC)、N-hydroxysuccinimide (NHS)爲媒合劑固定幾丁聚醣,之後測試幾丁聚醣表面之血栓形成與抗菌度。實驗中利用減弱全反射紅外線光譜儀及X光光電子能譜儀進行材料表面結構的分析,結果顯示:接枝丙烯酸的羧基(O=C-OH)與幾丁聚醣的胺基(-NH2)形成共價鍵結的醯胺鍵結(O=C-NH);而固定幾丁聚醣本身的C=O及NH2官能基均可被辨識。固定的幾丁聚醣經血液凝固測試顯示仍保有原來之生物特性,如:對血球產生聚集的效應。由纖維製品的定量抗菌性試驗結果表示,所固定的幾丁聚醣具有殺菌、抑菌的功能。因此,經固定幾丁聚醣的纖維將有潛力應用在拋棄式創傷敷材方面。

Low-temperature high-density O2 microwave plasma was utilized for the activation of non-woven polypropylene (PP) fabric. With several seconds of plasma treatment, highly oxidative groups were created at surface. The diluted acrylic acid (AAc) was subsequently graft-copolymerized to obtain a surface with functional and hydrophilic structure. To generate biocompatible function on the modified PP surface, chitosan molecules were then immobilized on the fabric by using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS) coupling agents. The chitosan-immobilized samples were subsequently tested for blood clot and antibacterial ability. Fourier-Transformed Infrared with Attenuated Total Reflection and X-ray Photoelectron Spectroscopy were utilized to identify the chemical structures at surface. Analytical result demonstrated that the O=C-OH group of the grafted pAAc was covalently bonded with chitosan, forming an amide bond (O=C-NH). The functional groups, C=O and NH2 of chitosan, were also well identified. Using blood clot tests, the immobilized chitosan still retained its original bioactivities, for example, it enhances red blood cells accumulation. From the quantitative antibacterial tests for textile product, the surface of chitosan-immobilized PP non-woven fabric provided hygienic function and inhibition of bacterial proliferation. Consequently, the chitosan-immobilized fabric is potentially applicable to disposal wound dressing.
