金門榕及廣東榕成年枝條之莖頂,在以MS培養基成份減去半量氮素的修正MS配方為基本,另添加Benzyl Adenine(BA)濃度為0.5-10.0 mg/l的半固體洋菜培養基上培養,之後莖頂有一段時間呈現生長停滯現象,莖頂外包的苞片逐漸由綠色轉為黑褐色,其間約為4個月,部份莖頂褐化死亡,部份莖頂則苞片脫離,成功的長出叢生狀不定芽體。BA濃度愈高,則莖頂保持高存活率的時間愈久,培植成功率亦隨著BA濃度的增高而昇高,當BA為10.0 mg/l時,金門榕的培植成功率為95.0%,廣東榕則為93.3%;但當BA為0.5 mg/l時,金門榕的培植成功率為22.2%,廣東榕為25.0%。
Shoot tips of adult Ficus plants, F. retusa cv. 'Kinmen' and cv. 'Kuntong', were cultured on modified medium containing half strength of nitrogenous compounds of Murashige and Skoog's (1962) medium supplimented with Benzyl Adenine (BA, 0.5 1.0 1.5 5.0 10.0 mg/l). After inoculation, the shoot tips stopped growing for about 4 months, and the surrounding bracts turned brown gradually. Part of the shoot tips turned brown then died, yet part of them successfully regenerated multiple adventitious buds. Shoot tips maintained high survival rate for a longer period at media with higher concentration of BA. Higher level of BA also caused higher regeneration rate, i.e. at 10 mg/l level, the regeneration rate of 'Kinmen' was 95.0% and 'Kuntong' was 93.3%; but at 0.5 mg/l level 'Kinmen' was 22.2% and 'Kuntong' was 25.0%.
